A surpreendente capacidade adaptativa bacteriana, em especial de algumas espécies patogênicas, tais como Vibrio cholerea e Pseudomonas aeruginosa, deve-se, em grande extensão, à multiplicidade e variedade de sistemas de sinalização capazes de sentir os mais diversos estímulos ambientais. Em especial, as vias de sinalização mediadas por c-di-GMP desempenham papel central na formação de biofilmes bacterianos. O trabalho apresentado nessa tese revela detalhes dos mecanismos moleculares de modulação da formação de biofilmes de P. aeruginosa através da regulação mediada por c-di-GMP nos níveis transcricional e pós-traducional. Apresentaremos as formas como c-di-GMP controla a transcrição do operon pel, via o fator de transcrição FleQ, e a síntese do exopolissacarídeo sintetizado e secretado pelo complexo proteico codificado nesse operon, através da ativação do efetor PelD. Nas considerações finais, as perspectivas de estudos futuros são brevemente discutidas à luz dos resultados obtidos.

Abstract in English
The surprising bacterial adaptive capacity, especially of some pathogenic species, such as Vibrio cholerae and Pseudomonas aeruginosa, is largely due to the multiplicity and variety of signaling systems sensing the most diverse environmental stimuli. C-di-GMP-mediated signaling pathways play a central role in the formation of bacterial biofilms. The work presented in this thesis reveals details of the molecular mechanisms underlaying de the formation of P. aeruginosa biofilm through c-di-GMP-mediate regulation at the transcriptional and post-translational levels. We will present the mechanism in which c-di-GMP controls the transcription of the operon pel, via the transcription factor FleQ, and the synthesis of exopolysaccharide synthesized and secreted by the protein complex encoded in this operon, through the activation of the PelD effector. Perspectives of future studies are briefly discussed in the light of the results obtained.
