Os receptores adrenérgicos formam a interface entre as catecolaminas endógenas e as células alvo para mediar os efeitos biológicos do sistema ervoso simpático (SNS). Até o momento há nove subtipos de receptores adrenérgicos clonados, dentre eles os subtipos 'alfa2A','alfa2B' e 'alfa2C' adrenérgicos (ARs). Os subtipos'alfa2AAR' e alfa2CAR' apresentam uma distribuição abrangente no sistema nervoso central e co-localizam-se em algumas regiões, podendo ser pré ou pós-sinápticos. O subtipo dominante no sistema nervoso central é o 'alfa2AAR' encontrado em regiões que contém corpos celulares de neurônios noradrenégicos, o que faz deste subtipo um importante mediador da auto-inibição pré sináptica da liberação de noradrenalina. Nos primeiros dois estudos apresentados na presente tese foram demonstrados que:1) os receptores 'alfa2AAR' e 'alfa2CAR' modulam o fluxo simpático por meio da inibição pós-sináptica de neurônios pré-motores simpáticos no bulbo rosto-ventro-lateral (principal subtipo 'alfa2AAR') e funcionam como autorreceptores inibitórios nos terminais nervosos simpáticos. Quando ativados por noradrenalina inibem o influxo de 'Ca2+' em cultura primária de neurônios simpáticos (o efeito auto-inibitório principal é do subtipo 'alfa2AAR' e é potencializado pela ativação do subtipo 'alfa2CARKO'). Cabe ressaltar que a função desses subtipos de receptores 'alfa2ARs' depende de sua localização subcelular em neurônios simpáticos em cultura e o tempo de cultivo é uma variável importante a ser levada em consideração nos experimentos que abordam a função desses autorreceptores in vitro. 2) com a deleção gênica dos receptores 'alfa2AAR' e o 'alfa2CAR'instala-se um quadro de hiperatividade simpática em camundongos. Essa hiperatividade simpática é acompanhada de disfunção ventricular progressiva e de remodelamento cardíaco com aumento xpressivo nas concentrações cardíacas de Ang II que ulminam em edema pulmonar e aumento na taxa de mortalidade aos sete meses de idade

Adrenergic receptors form the interface between endogenous catecholamines and target cells to mediate the biological effects of the sympathetic nervous system (SNS). So far there are nine cloned adrenergic receptor subtypes, among them the 'alpha2A', 'alpha2B' and 'alpha2C' adrenergic subtypes (ARs). The 'alpha2AAR' and alpha2CAR' subtypes have a wide distribution in the central nervous system and co-localize in some regions, and may be pre- or postsynaptic. The dominant subtype in the central nervous system is the 'alpha2AAR' found in regions containing cell bodies of noradrenergic neurons, which makes this subtype an important mediator of presynaptic self-inhibition of noradrenaline release. In the first two studies presented in this thesis, it was demonstrated that: 1) 'alpha2AAR' and 'alpha2CAR' receptors modulate sympathetic outflow through postsynaptic inhibition of sympathetic premotor neurons in the face-ventro-lateral medulla (main subtype 'alpha2AAR') and function as inhibitory autoreceptors on sympathetic nerve terminals. When activated by noradrenaline, they inhibit the influx of 'Ca2+' in primary cultured sympathetic neurons (the main auto-inhibitory effect is of the 'alpha2AAR' subtype and is potentiated by the activation of the 'alpha2CARKO' subtype). It should be noted that the function of these 'alpha2ARs' receptor subtypes depends on their subcellular location in sympathetic neurons in culture, and the culture time is an important variable to be taken into account in experiments that address the function of these autoreceptors in vitro. 2) with the gene deletion of the 'alpha2AAR' and 'alpha2CAR' receptors, a picture of sympathetic hyperactivity is installed in mice. This sympathetic hyperactivity is accompanied by progressive ventricular dysfunction and cardiac remodeling with a significant increase in cardiac Ang II concentrations that culminate in pulmonary edema and an increase in the mortality rate at seven months of age
