Cianobactérias são organismos fotossintetizantes que habitam diferentes ecossistemas. No ambiente de água doce, especialmente em represas, lagos e lagoas, podem crescer de forma acelerada (floração) e causar problemas de toxicidade e contaminação da água. Elas produzem diversos metabólitos que podem ser tóxicos, como microcistinas e nodularinas, mais amplamente conhecidas e estudadas. No entanto, a riqueza de compostos produzidos é alta e ainda muito pouco se conhece em relação à sua estrutura química e sua toxicidade. Sabe-se, entretanto, que boa parte destes compostos são de origem peptídica, geralmente polipeptídeos necessários para sua sobrevivência. Desta forma, esse estudo teve como objetivo o desenvolvimento de uma metodologia analítica por meio de cromatografia líquida de alta eficiência (em inglês, High-Performance Liquid Chromatography, HPLC ou LC) acoplada a espectrometria de massas (em inglês, mass spectrometry, MS) para a determinação quantitativa e simultânea dos cianopeptídeos namalídeos e espumiginas. Ambos compostos foram recém descobertos, e analisados neste estudo a partir da cianobactéria Sphaerospermopsis torques-reginae (cepa ITEP- 024). Foram testados diversos métodos de extração, purificação e separação com sua biomassa seca. Todos com vantagens e desvantagens, no entanto, a melhor opção foi uma extração direta. Outro ponto avaliado foram os padrões internos (PI), na qual foram testados diversos compostos quimicamente similares aos do estudo, e alguns se mostraram promissores para o estudo, como o HHL (N-Hippuryl-His-Leu-hydratade) e GPA (Z-Gly-Pro-Arg-p-nitroanilida acetate salt). O método cromatográfico foi otimizado para as análises.

Cyanobacteria are photosynthetic organisms that inhabit different ecosystems. In the freshwater environment, especially in dams, lakes, and ponds, they can proliferate (bloom) and cause problems of toxicity and water contamination. They produce several metabolites that can be toxic, such as microcystins and nodularins, which are more widely known and studied. However, the compounds' richness is high and very little is known about their chemical structure and toxicity. It is known, however, that most of these compounds are of peptide origin, generally polypeptides necessary for their survival. Thus, this study aimed to develop an analytical methodology using High-Performance Liquid Chromatography (HPLC or LC) coupled with mass spectrometry (MS) for the quantitative and simultaneous determination of namalide and spumigin cyanopeptides. Both compounds were recently discovered and analyzed in this study from the cyanobacterium Sphaerospermopsis torques-reginae (strain ITEP-024). Several extractions, purification, and separation methods were tested with their dry biomass. All with advantages and disadvantages. However, the best option was a direct extraction. Another point evaluated was the internal standards (IP), in which several compounds chemically similar to those in the study were tested, and some showed promise for the study, such as HHL (N-Hippuryl-His-Leu-hydrated) and GPA (Z-Gly-Pro-Arg-pnitroanilida acetate salt). The chromatographic method was optimized for the analyses.
