Nos últimos anos, a população idosa tem aumentado, consideravelmente, no cenário mundial. Tais pessoas tendem a apresentar alterações nutricionais, ocasionadas pelos mais diversos mecanismos. Ambos os processos, envelhecimento e comprometimento nutricional, podem afetar diversas funções do organismo, por exemplo, o sistema imunológico. Por esta razão, o presente estudo tem por objetivo avaliar determinados aspectos relacionados à resposta imunológica inata e seus mecanismos, em um modelo animal idoso, submetido à carência proteica. Para isso, foram utilizados camundongos machos, de linhagem C57BL/6, divididos em dois grupos relacionados à idade: grupo de animais jovens (3-6 meses) e outro de animais idosos (18-19 meses) que, após alcançarem a idade desejada, foram separados em dois grupos, controle e desnutrido. Ao longo do processo, foram realizados mapeamento nutricional, composição corporal, análise bioquímica e hematológica. Foram coletadas células da cavidade peritoneal e realizados imunofenotipagem, além de teste de espraiamento, fagocitose, atividade fungicida com Candida albicans, bem como produção de citocinas e avaliação da expressão dos fatores de transcrição NFκB e STAT 3, após estímulo com LPS. Os animais, desnutridos jovens e idosos, apresentaram anemia e leucopenia, bem como níveis menores de proteína total e albumina. A capacidade de espraiamento, fagocitose e atividade fungicida apresentou-se reduzida nas células dos animais jovens desnutridos, bem como nas células dos animais idosos, independente do estado nutricional. Ainda, animais idosos apresentaram menor porcentagem de células F4/80+/CD11b+ e menor expressão de CD86+, independente do estado nutricional. Porém, observamos redução na expressão de TLR4+ somente nos animais desnutridos, independentemente da idade. Em relação à produção de citocinas, observamos que células de animais desnutridos, tanto do grupo jovem como do idoso, apresentaram menor capacidade de produção de IL-1α e TNF-α. Também observamos redução na produção de TNF-α em células de animais idosos controle. Em relação à produção de IL-10, observamos aumento no grupo idoso, principalmente, nos animais desnutridos. Ao analisar a expressão da relação do fator de transcrição NFkB fosforilado pela sua fração total, observamos que células de animais desnutridos apresentaram redução, independentemente da idade. Dessa forma, tais resultados permitem concluir que o envelhecimento e a carência proteica comprometem a resposta imune celular e, quando associados, esse comprometimento tende a ser mais acentuado.

The elderly population has increased considerably in recent years on the world stage. This population tend to have nutritional changes, caused by the most diverse mechanisms. Both processes, aging and nutritional impairment, can affect various functions of the body, such as the immune system. Therefore, this study aims to evaluate some aspects related to the innate immune response and its mechanisms, in an elderly animal model submitted to protein deficiency. For this, C57BL / 6 male mice were used, divided into two age-related groups: group of young animals (3-6 months) and another group of elderly animals (18-19 months) that, after reaching the desired age, were separated into two groups: control and malnourished. Throughout the process, nutritional mapping, body composition, biochemical and hematological analysis were carried out. The cells of the peritoneal cavity were collected and immunophenotyping, spreading test, phagocytosis and fungicidal activity with Candida albicans were performed, as well as cytokine production and evaluation of the expression of the transcription factors NFκB and STAT 3, after stimulation with LPS. The animals, malnourished young and old, had anemia and leukopenia, as well as lower levels of total protein and albumin. Spreading capacity, phagocytosis and fungicidal activity were reduced in the cells of malnourished young animals, as well as in the cells of elderly animals, regardless of nutritional status. Additionally, elderly animals showed a lower percentage of F4/80 + / CD11b + cells and a lower expression of CD86 +, regardless of nutritional status, but we observed a reduction in the expression of TLR4 + only in malnourished animals, regardless of age. Regarding the production of cytokines, we observed that cells from malnourished animals, both from the young and the elderly group, had a lower production capacity of IL-1α and TNF-α, as well as a reduction in the production of TNF-α in cells of elderly animals. control. Additionally, in relation to the production of IL-10, we observed an increase in the elderly group, mainly in malnourished animals. When analyzing the expression of the ratio of the transcription factor NF?B phosphorylated by its total fraction, we observed that cells from malnourished animals showed a reduction regardless of age. Thus, these results allow us to conclude that aging and protein deficiency compromise the cellular immune response and that when associated, this impairment tends to be more pronounced.
