Ganho de peso corporal é uma dos efeitos indesejáveis associados aos antipsicóticos, de importância para a saúde global dos pacientes, bem como para sua adesão ao tratamento. Sendo assim, investigaram-se a relação entre o ganho de peso durante o tratamento com olanzapina e haloperidol e o impacto nos níveis séricos de leptina e de neuropeptídeo Y em portadores de esquizofrenia. Participaram do estudo 34 pacientes do sexo masculino, 18 em uso de olanzapina (5-20 mg/dia) e 16 de haloperidol (5-20 mg/dia), ambos em regime de monoterapia. Os resultados indicaram que: (a) tanto a olanzapina como o haloperidol induziram graus de incremento do peso clinicamente significantes e que (b) o grau de aquisição do peso correlacionou-se positivamente com o incremento da leptina e do neuropeptídeo Y. Estes resultados sugerem que os antipsicóticos interferem na via de sinalização da leptina e do neuropeptídeo Y, prejudicando os efeitos dos mesmos no controle do comportamento alimentar.

Body weight gain is a frequent side effect of most antipsychotic drugs, which impairs health and interferes with treatment compliance. We investigated weight gain during antipsychotic treatment with olanzapine and haloperidol and the relationship between leptin and neuropeptide Y serum levels in schizophrenic patients. Thirty-four male schizophrenic patients were evaluated, 18 who received orally olanzapine (5-20 mg/day) and 16 haloperidol (5-20 mg/day), both in monotherapy. The results indicated that: (a) olanzapine and haloperidol were both associated with significant increases in body weight and (b) there was a positive correlation between increments in weight and in both leptin and neuropeptide Y serum levels. These results suggest that antipsychotic drugs disrupt the leptin and the neuropeptide Y pathways and consequently impair the effects of these mediators upon the control of feeding behavior.
