A busca por alternativas terapêuticas para o tratamento da doença de Chagas é de grande importância, pois, atualmente, existem apenas dois fármacos disponíveis no mercado, o benznidazol e o nifurtimox, que apresentam efeitos adversos consideráveis. Compostos 5-nitroheterocíclicos com atividade antiparasitária têm apresentado resultados encorajadores para o tratamento desta doença. Devido à preferência pela administração oral para o tratamento de doenças crônicas, estudar a permeabilidade de uma nova molécula é fundamental. O objetivo do trabalho foi avaliar a citotoxicidade e a permeabilidade de análogos da nifuroxazida (compostos 22, 23 e 24) com atividade frente ao T. cruzi, utilizando ensaios in vitro com monocamadas de células Caco-2, modelo in vitro mais empregado para a predição da permeabilidade e absorção in vivo. Realizaram-se testes de citotoxicidade destes compostos, empregando o ensaio MTT (brometo de 3-(4,5-dimetiltiazol-2-il)2,5-difenil tetrazólio) para avaliar a viabilidade celular e determinar a concentração a ser utilizada nos experimentos de permeabilidade. Para a quantificação destas moléculas foi desenvolvido e validado um método bioanalítico de acordo com a legislação vigente. Obteve-se a viabilidade celular de até 70% nas concentrações de 9,3; 6,7 e 3,1 µg/mL (n=3) para os compostos 22, 23 e 24, respectivamente. O método desenvolvido demonstrou seletividade, linearidade, precisão, exatidão e estabilidade, tornando-o aplicável à quantificação dos compostos em estudo. Na avaliação da permeabilidade aparente (Papp), os padrões de alta (metoprolol) e baixa (hidroclorotiazida) apresentaram valores de Papp iguais a 29,4 e 0,54 x 10-6 cm/s, respectivamente. Para os análogos à nifuroxazida (C22, C23 e C24) foram obtidos os valores de 58,52; 53,97 e 46,07 x 10-6 cm/s, respectivamente, o que os classifica como compostos de alta permeabilidade. Os resultados são promissores e importantes para encorajar a continuidade da pesquisa com os candidatos a fármacos avaliados, colaborando com a obtenção de alternativas para o tratamento da referida doença, considerada uma doença negligenciada no Brasil.

The search for therapeutic alternatives for the treatment of Chagas' disease has great importance, since that currently there are only two drugs available on the market: benznidazol and nifurtimox, which have considerable adverse effects. 5- nitroheterocyclic compounds with antiparasitic activity have shown encouraging results for the treatment of this disease. Due to the preference for oral administration for the treatment of chronic diseases, studying the permeability of a new molecule is essential. The objective of this study was to evaluate the cytotoxicity and permeability of nifuroxazide analogues (compounds 22, 23 and 24) with activity against T. cruzi, using in vitro assays with Caco-2 cell monolayers, the most used in vitro model for the prediction of permeability and in vivo absorption. Cytotoxicity tests of these compounds were performed, using the MTT assay (3-(4,5-dimethylthiazol-2-yl) 2,5-diphenyl tetrazolium bromide) to assess cell viability and determine the concentration to be used in the experiments permeability. For the quantification of these molecules, a bioanalytical method was developed and validated according to the current legislation. Cell viability of up to 70% was obtained at concentrations of 9.3; 6.7 and 3.1 µg/mL (n=3) for compounds 22, 23 and 24, respectively. The developed method demonstrated selectivity, linearity, precision, accuracy and stability, making it applicable to the quantification of the compounds under study. In the assessment of apparent permeability (Papp), the high (metoprolol) and low (hydrochlorothiazide) patterns showed Papp values equal to 29.4 and 0.54 x 10-6 cm/s, respectively. For analogues to nifuroxazide (C22, C23 and C24) the values of 58.52 were obtained; 53.97 and 46.07 x 10-6 cm/s, respectively, which classifies them as compounds with high permeability. The results are promising and important to encourage the continuity of the research with the candidates of evaluated drugs, collaborating with the obtaining of alternatives for the treatment of the referred disease, considered a neglected one in Brazil.
