Neutrófilos polimorfonucleares (PMN) têm um importante papel na defesa do hospedeiro contra infecções. Estas células produzem grande quantidade de espécies reativas de oxigênio (especialmente, O2·- e H2O2) com atividade microbicida. Muitos estudos in vitro têm sido usados para estimar a produção de espécies reativas in vivo. Nestes estudos PMN isolados de sangue periférico por gradientes de densidade (gradiente de Ficoll-Hypaque e sedimentação em dextran) são comumente usados. Este método resulta na obtenção de alta homogeneidade celular, entretanto, leva a prévia ativação celular devido a perturbações na membrana. Neste trabalho foi comparada a produção de espécies reativas de oxigênio por PMN obtidos por gradiente de densidade (1x105cels/ml) com PMN obtidos por sedimentação espontânea (2x105cels/ml) ou presentes no sangue sem qualquer manipulação. Embora o processo de sedimentação espontânea não leve a uma população celular homogênea, é suposto que preserve as células de estimulação durante o isolamento. A produção de O2·- e a ativação do sistema H2O2/MPO foi monitorada respectivamente por quimluminescência dependente de lucigenina (2,5x105M) e luminol (1x105M). A produção de O2·- foi também monitorada pela redução do citocromo c. A produção de O2·- e a ativação do sistema H2O2/MPO foram maiores para as células isoladas por gradiente de densidade com ou sem qualquer estimulação (2,5 mg/ml de zymosan opsonizado, 50 ng/ml de acetato de forbol miristato e 4x106M de formilmetionil-leucil-fenilalanina). Adicionalmente a liberação de MPO foi significantemente maior para as células obtidas por gradiente de densidade quando comparadas às células obtidas por sedimentação espontânea. Os amplificadores clássicos da reação HRP/H2O2/luminol parecem ser ineficientes para as reações do sistema MPO/H2O2/luminol. A razão para esta ausência de amplificação deverá ser elucidada em estudos posteriores. Este trabalho mostra claramente que os processos de obtenção de PMN devem afetar respostas celulares importantes, contribuindo para alguns erros de interpretação de resultados obtidos in vitro.

Polymorphonuclear neutrophils (PMN) have an important role in the host defense response to infection. These cells produce large amounts of reactive oxygen species (O2·- and H2O2) with microbicidal activity. Several in vitro studies have been used in order to estimate the yield of reactive species in vivo. In these studies PMN isolated from peripheral blood by density gradients (Ficoll-Hypaque gradient and dextran sedimentation) are commonly used. This method yield a high homogeneity in cellular population whereas leads to prior cellular activation due to membrane perturbation. ln this work it was compared the reactive oxygen production of PMN isolated by density gradient (1x105 cells/mL) with PMN obtained by spontaneous sedimentation (2.5x105 cells/mL) or present in blood withou any manipulation. Although spontaneous sedimentation does not lead to a homogeneous cellular population it is suppose to preserve the cells from spurious stimulus during isolation. The production of O2·- and the evaluation of the H2O2/mieloperoxidase (MPO) system was followed by lucigenin (2.5x10-5M) and luminol (1x10-5M) enhanced chemiluminescence, respectively. O2·- production was also followed by cytochrome e reduction. Both, O2·- and MPO/H2O2 system, were higher for cells isolated by density with or without any added stimuli (2.5 mg/ml opsonized zymosan, 50 ng/mL phorbol myristate acetate and 4x10-6M N-formyl-methionyl-leucyl-phenylalanine). Furthermore, the release of MPO was significantly higher for cells obtained by density gradient when compareci to cells obtained by spontaneous sedimentation. The classical enhancers of the HRP/H2O2/luminol reaction seems to be ineffective for the MPO/H2O2/luminol reaction. The failure of this enhancement haven't known and may be elucidate in future studies. This work clearly show that the PMN isolation procedure might affect important cellular responses contributing to some misinterpret in vitro results.
