A alta característica epidêmica e a mortalidade fazem da doença meningocócica um sério problema de saúde pública. Isto fez com que vários centros de pesquisas direcionassem sua atenção para o desenvolvimento de uma vacina contra o meningococo B. As vacinas existentes no momento são contra os sorogrupos A, C, Y e W135. Para conseguir uma vacina segura muitos antígenos da membrana externa bacteriana tomaram-se alvo de estudos. Um dos Componentes que tem sido estudado para ser utilizado em uma preparação vacinai contra Neisseria menigitidis B é o lipopolissacáride (LPS). Lipopolissacárides são macrornotéculas lipoprotêicas que fazem parte da membrana externa das bactérias gram-negativas. O interessante é que muitos estudos com estas moléculas mostraram características adjuvantes na resposta imune contra a N. meningitidis B. Entretanto um problema com estas moléculas é o efeito tóxico que elas causam devido ao lipídio A contido em sua estrutura. O presente estudo teve como finalidade a avaliação da especificidade e capacidade protetora dos anticorpos induzidos em modelo animal com outer membrane components OMC de cepas de N. meningitidis B:4:P1.15 selecionadas por meio de "colony-blot" utilizando anticorpos monoclonais anti-L3, 7 ,9 e L8 para maior e menor expressão dos LPS dos imunotipos L3,7,9 e L8. A princípio realizamos a seleção das cepas por "colony-blot" para depois procedermos à extração dos componentes de membrana externa bacteriana(OMC) obtidos das cepas selecionadas (B:4:P1.15 L3,7,9↑, B:4:P1.15 L3,7,9↓, B:4:P1.15 L8↑ e B:4:P1.15 L8-↓) e à imunização pela via intranasal (IN) e intramuscular (IM) dos camundongos BALB/c. Nesse estudo foram analisadas: a reatividade homóloga de anticorpos dos isótipos lgM, lgG e lgA presentes nos soros e nas salivas dos camundongos imunizados pelas vias IN e IM e a presença no soro de anticorpos com atividade bactericida. Nos soros de camundongos imunizados pela via IN foram analisadas também a reatividade de anticorpos dos isótipos lgM, lgG e lgA presentes 4 meses e 1 ano após a imunização e a reatividade de anticorpos dos isótipos lgM, lgG e lgA de reatividade cruzada contra suspensão de células bacterianas íntegras das cepas de N. meningitidis heterólogas (B:4:P1.9,B:4:NT, B:14:NT, e C:2a:P1.2). Na análise da presença de anticorpos no soro (obtidos em diferentes dias após a imunização) e na saliva, foi observada a resposta tanto contra suspensão de células bacterianas íntegras como para OMC das cepas selecionadas. Os resultados da dosagem de anticorpos por ELISA mostram que os camundongos imunizados pela via IM produziram altos títulos de anticorpos dos isótipos lgG e os camundongos imunizados pela via IN produziram anticorpos dos isótipos lgG e lgA específicos contra células bacterianas íntegras das cepas homólogas e heterólogas de N. meningitidis. A análise da especificidade por "Immunoblotting" dos anticorpos presentes nos soros dos camundongos imunizados pela via IN e IM mostrou que a resposta foi direcionada para proteínas da classe 1,2, e 5 e também para proteínas de 50, 65 e 70 kDa da suspensão de células bacterianas integras e OMC da cepas B:4:P1.15 L3,7,9↑, B:4:P1 .15 L3,7,9↓, B:4:P1.15 L8↑ e B:4:P1.15 L8↓. A especificidade dos anticorpos dos isótipos lgG e lgA (obtidos soros de camundongos imunizados pela via IN) contra cepas B:4:P1.9, B:14:NT, B:4:NT e C:2a:P1.2 mostrou que a resposta foi direcionada principalmente contra proteínas acima de 50 kDa. Na saliva de camundongos imunizados pela via IN, anticorpos do isótipo lgA foram observados por meio de ELISA e "Immunoblotting", sendo que por meio de "Immunoblotting", verificamos que esta resposta foi direcionada para proteínas de 28 kDa, 33 kDa e acima de 50 kDa. Os resultados apresentados dos ensaios bactericidas mostraram que os grupos de camundongos que receberam imunização pela via IN produziram altos títulos de anticorpos com atividade bactericida contra as cepas homólogas. Quando comparamos as preparações antigênicas obtidas de cepas com maior ou menor expressão dos inunotipos L3,7,9 e L8 não pudemos observar diferenças significativas por meio de ELISA. Entretanto por meio de "Immunoblotting" observamos diferenças na especificidade da resposta, especialmente para peptídeos de alto peso molecular. As análises da especificidade dos anticorpos indicam que a seleção de cepas com maior expressão de LPS pode ser importante para iniciar uma resposta efetiva tanto sistêmica como de mucosa quando apresentados em uma preparação vacinai através da via nasal e vem ressaltar a importância do estabelecimento do imunotipo na produção de vacina meningocócica capaz de induzir uma imunidade duradoura em todas as idades tanto pela via mucosa como sistêmica.

Meningococcal disease poses a serious public health problem, due to their characteristics of being highly epidemic, with a high mortality rate. This fact led many research centers to tum their efforts to the development of a vaccine against Neisseria meningitides serogroup B. The vaccines available at the present time are directed to serogroups A, C, Y and W135. In order to obtain a safe vaccine, many of the antigens of the outer membrane of the bacteria are being studied. Among the components being studied for eventual use in a vaccine against N. meningitides B are the lipopolysaccharides (LPS). Lipopolysaccharides are complex macromolecules lipoprotein which integrate the outer membrane of gram-negative bacteria. It is interesting that many of the studies involving these molecules have shown adjuvant characteristics in the immune response against N. meningitides B. However, there is a problem with these molecules, due to the toxic effects of the lipid A present in their structure. The present study had the aim of evaluating the specificity and protective activity of the antibodies induced in an animal model by the outer membrane complex (OMC) obtained from strains of N. meningitides B:4:P1.15. The strains used for immunization of mice were selected through colony-blot, using anti L3,7,9 and L8 monoclonal antibodies, for the highest and the lowest expression of the LPS of immunotypes L3,7,9 and L8. First we selected the strains using colony blots, then we performed the extraction of the OMC of the bacteria belonging to the selected strains (B:4:P1:15 L3, 7, 9↑, B:4:P1.15 L3, 7, 9↓, B:4:P1:15 L8↑ and B:4:P1:15 L8↓) followed by, the intranasal (IN) and intramuscular (IM) immunization of the BALB/c mice with the purified OMC. The present study analyzed the reactivity of the antibodies (lgM, lgG and lgA) found in the serum and in the saliva of mice who were immunized IN or IM, as well as the presence in the serum of antibodies with bactericidal activity. In the serum of the IN immunized mice, antibodies found four months and one year after immunization and with cross-reactivity against a whole-cell suspension of heterologous strains of N. meningitides bacteria (B:4:P1.9, B:4:NT, B:14:NT and C:2a:P1.2) were also tested. The analysis of the presence of antibodies in the serum (obtained 30 after immunization) and in the saliva showed reactivity against both the whole-cell bacteria and the OMC of the strains selected. In study of antibodies found four months and one year after the immunization against the strains B:4:P1.9, B:4:NT, B:14:NT and C2a:P1.2 only the reactivity against the whole-cell suspension of the bacteria were analyzed. The results of the ELISA test shows that IM immunized mice produced higher levels of antibodies of the lgG isotype and the IN immunized ones produced higher levels of antibodies of the lgG and lgA isotypes, specific against whole-cells of homologous and heterologous strains of N. meningitides. The analysis of specificity through the Immunoblotting of the antibodies found in the serum of the IM and IN immunized mice revealed that the response was directed mainly against the proteins of classes 1, 2 and 5, and also against proteins 50, 65 and 70 kDa of the whole-cell suspension bacteria and the OMC of strains B:4:P1:15 L3, 7, 9↑, B:4:P1 :15 L3, 7, 9↓, B:4:P1 .15 L8↑ and B:4:P1:15 L8↓. The specificity of antibodies of isotypes lgG and lgA (obtained from the serum of intranasally immunized mice) against strains B:4:P1.9, B:14NT, 8:4:NT and C.2a.P1.2 revealed that the response was directed mainly against proteins of molecular weight higher than 50 kDa. In the saliva of IN immunized mice the reaction of antibodies of the lgA isotype was also observed by ELISA and Immunoblotting. The response was directed against proteins of 28 kDa, 33 kDa and higher than 50 kDa. When the antigen preparations obtained from the strains with the highest and lowest expressions of immunotypes L3,7,9 and L8 were compared, no significant differences were found by ELISA. Immunoblotting analysis, however, showed differences in the specificity of the response, particularly for peptides of high molecular mass. The titers obtained by the assay of bactericidal assay were higher for the mice IN immunized with strains selected for LPS L3, 7, 9 and L8. The results of antibodie's specificity indicate that the selection of LPS may be important to elicit systemic as well as local muccosal effective immune response, when used in an intranasal vaccine preparation. This work indicates the importance of selecting LPS immunotypes in order to obtain a meningococcal vaccine capable of inducing a lasting immunity in people of all ages, in both the muccosal and the systemic modes of admission.
