Introdução: A doença de Alzheimer (DA) é uma patologia neurodegenerativa progressiva e fatal que induz deterioração cognitiva e da memória. A injeção intracerebroventricular de estreptozotocina (icv-STZ) demonstrou ser um modelo experimental da doença de Alzheimer esporádica em roedores, reproduzindo muitas das características comportamentais e moleculares dessa doença. A geleia real (GR) é uma substância secretada pelas glândulas hipofaríngeanas e mandibulares de abelhas operárias da espécie Apis mellifera. Objetivos: Investigar os efeitos do consumo oral e prolongado da GR em ratos submetidos ao modelo icv-STZ na aprendizagem e na memória pelo teste do labirinto aquático de Morris (MWM), teste do Reconhecimento de Objetos (RO), atividade geral em Campo Aberto, análises histológicas caracterizando a atividade de células gliais e ensaios de imunoprecipitação com foco na expressão de subtipos de receptores muscarínicos de acetilcolina (mAChRs) (M1-M5) no hipocampo de ratos. Métodos: Ratos Wistar machos adultos foram divididos em quatro grupos (CTR, STZ, GR, STZ-GR). A administração bilateral de estreptozotocina (STZ) (3 mg/kg) ou solução Ringer foi realizada por estereotaxia. Após 7 dias da cirurgia, os animais receberam diariamente GR (200 mg/kg) ou solução controle por gavagem por 14 dias consecutivos. O teste do labirinto aquático de Morris foi realizado antes (P1), após (P2) a injeção de icv-STZ, e durante a administração da GR (P3). O teste RO foi realizado no 14° dia após a cirurgia icv-STZ. O teste de Campo Aberto foi realizado no 13° dia após a cirurgia icv-STZ. No 21° dia os animais foram sacrificados e a análise da glia reativa no hipocampo foi feita, assim como a expressão de cada subtipo de receptor muscarínico por ensaios de imunoprecipitação com anticorpo que reconhece cada um dos subtipos de mAChRs. Resultados: No Período P2: icv-STZ induziu um aumento na latência, distância percorrida e uma diminuição na % Tempo no contador. No período P3, o consumo prolongado da GR melhorou o aprendizado espacial dos animais do grupo icv-STZ, conforme observado pela redução da latência em T2, T3 e T4. Teste RO: Os ratos tratados com STZ passaram menos tempo explorando o novo objeto do que o objeto familiar quando comparados aos grupos CTR e GR, após um período de 1 hora e 24 horas. Atividade Geral em Campo Aberto: a injeção icv-STZ e o tratamento prolongado com GR não causaram qualquer alteração nos parâmetros investigados. O tratamento com GR produziu uma redução da astrogliose induzida por STZ nas regiões CA1, CA3, GD e HL do hipocampo de ratos. Os sujeitos tratados com STZ (grupo STZ) apresentaram uma diminuição da expressão do subtipo M1 e o tratamento com GR (grupo STZ-GR) recuperou a expressão. Entretanto, os subtipos M2, M3, M4 e M5 não diferiram entre os grupos. Nossos resultados mostram que a injeção icv-STZ causou um prejuízo cognitivo, induziu um aumento da expressão de células gliais hipocampais e uma redução da expressão do subtipo de mAChRs M1. Por outro lado, o tratamento prolongado com a GR reverteu parcialmente os prejuízos cognitivos, reduziu a densidade de células gliais e recuperou a expressão do receptor M1 comparativamente ao grupo STZ.

Introduction: Alzheimer's disease (AD) is a progressive and fatal neurodegenerative disorder that induces cognitive and memory deterioration. Intracerebroventricular injection of streptozotocin (icv-STZ) has been shown to be an experimental model of sporadic Alzheimer's disease in rodents, reproducing many of the behavioral and molecular characteristics of this disease. Royal jelly (RJ) is a substance secreted by the hypopharyngeal and mandibular glands of worker bees of the species Apis mellifera. Objectives: To investigate the effects of oral and long-term consumption of RJ on rats submitted to the icv-STZ model on learning and memory by the Morris Water Maze Test (MWM), object recognition (OR) test, general activity in open field test, and histological analyzes characterizing the activity of Glial cells besides immunoprecipitation assays focusing on Expression of each muscarinic acethylcholine receptor (mAChRs) subtypes (M1 to M5) in rat hippocampus. Methods: Adult male Wistar rats were divided into four groups (CTR, STZ, CTR-RJ, STZ-RJ). Stereotaxic surgery for bilateral icv administration of streptozotocin (STZ) (3 mg/kg) or control solution was performed. The animals received daily RJ (200 mg/kg) or control solution by gavage for 14 consecutive days. The MWM Task was performed before (P1), after (P2) the ICV-STZ injection, and during the RJ administration for eleven days (P3). The OR test was performed in the 14th day after icv-STZ surgery. The Open Field test was performed in the 13th day after the icv-STZ surgery. In the 21° day after surgery the animals were euthanized and the reactive glia in the hippocampus was analyzed, as well as expression of each mAChR subtype (M1 to M5) by immunoprecipitation assays. Results: Period P2: icv-STZ induced an increase in the latency to find the platform, distance (path length) and a decrease in the percentage of time spent within the critical counter, indicating a work memory disruption. In the P3 period, long-term consumption of RJ improved spatial learning rate was improved as observed by the reduction of latency in T2, T3 and T4. RO test: STZ-treated rats spent less time exploring the new object than the familiar object when compared to the CTR and CTR-GR groups, after 1 and 24 hours indicating that the STZ induced an impairment short-term memory and long-term memory and that the RJ did not revert this effect. Open Field: the icv-STZ injection and the prolonged treatment with GR did not cause any change in the parameters analysed. The long-term oral treatment with RJ induced significant effects reducing astrogliosis STZ-induced in the rat hippocampal CA3, GD, HL and CA1 regions. The animals injected with icv-STZ (STZ group) showed a decrease in the expression of M1 subtype and the long-term oral treatment with RJ (STZ-RJ) recovered the expression. However, the M2, M3, M4 and M5 subtypes did not differ among the groups. Our results show that the icv-STZ injection was able to cause cognitive impairment, induce increased expression of hippocampal Glial cells and induced a decrease in the expression of M1 mAChR subtype. In addition, prolonged treatment with GR was able to partially reverse cognitive impairments, reduce the density of glial cells and recovered the expression of M1 mAChR compared to STZ group.
