O Streptococcus agalactiae ou Streptococcus do Grupo B (GBS - group B Streptococcus) é um coco gram-positivo, caracterizado por seu potencial em causar doenças invasivas em recém-nascidos (RNs) como sepse, meningite e pneumonia. Diversos microrganismos patogênicos são capazes de expressar diferentes proteínas, as quais podem representar antígenos relevantes para indução de proteção no hospedeiro. A Proteína Transportadora de Poliaminas D (PotD), é uma proteína de superfície, conservada entre os diferentes sorotipos de GBS (Ia, Ib, II ao IX), pertencendo à uma grande família de transportadores ativos em diversas células. As poliaminas são fundamentais para a sobrevivência e crescimento da célula, exercendo funções metabólicas e fisiológicas. Vacinas de DNA possuem em sua formulação genes do patógeno que são responsáveis por codificar proteínas que podem conter epítopos antigênicos que conferem imunidade ao hospedeiro, resultando em uma proteção imunológica. Sendo assim, o objetivo deste trabalho é desenvolver uma abordagem vacinal baseada no uso de vacina de DNA, utilizando o gene potD para indução de resposta imune contra o patógeno. Através da clonagem, a construção vacinal pVAX-potD foi testada em cultura de células de mamíferos HEK293T. Os resultados obtidos até o momento sugerem que com apenas uma única dose da vacina de DNA, administrada em camundongos, utilizando eletroporação, o sistema imune foi capaz de reconhecer o antígeno alvo e gerar anticorpos específicos contra a bactéria.

Group B Streptococcus agalactiae (GBS) is a gram-positive coconut characterized by its potential to cause invasive diseases in newborns (NBs) such as sepsis, meningitis, and pneumonia. Several pathogenic microorganisms can express different proteins, which may represent antigens relevant to induction of protection in the host. Polyamine Carrier Protein D (PotD) is a surface protein, conserved among the different GBS serotypes (Ia, Ib, II to IX), belonging to a large family of active transporters in several cells. Polyamines are fundamental for cell survival and growth, performing metabolic and physiological functions. DNA vaccines have pathogen genes in their formulation that are responsible for encoding proteins that may contain antigenic epitopes that confer immunity to the host, resulting in immune protection. Thus, the objective of this work is to develop a vaccine approach based on the use of DNA vaccine, using the potD gene to induce immune response against the pathogen. Through cloning, the pVAX-potD vaccine construction was tested in HEK293T mammalian cell culture. The results obtained so far suggest that with only a single dose of the DNA vaccine, administered in mice, using electroporation, the immune system was able to recognize the target antigen and generate specific antibodies against the bacterium.
