Biofilmes dentais caracterizam um problema de saúde oral pois podem culminar no desenvolvimento de cáries e outras doenças bucais bastante onerosas. Seus principais constituintes são os polissacarídeos extracelulares insolúveis. Tais componentes estão diretamente relacionados com a cariogenicidade das comunidades bacterianas e são formados por ligações glicosídicas (1 → 3)- α e (1 → 6)- α.Considerando a estrutura química dessas moléculas e sua importância para o biofilme dental, o emprego de enzimas exógenas capazes de clivar essas ligações glicosídicas, denominadas glucanases, se mostra uma estratégia promissora para o controle desses biofilmes. No presente trabalho, uma (1 → 3)- α (PmGH87) e uma (1 → 6)-α (CoGH66) glucanase de Prevotella melaninogenica e Capnocytophaga ochraceae, respectivamente, foram clonadas e expressas heterologamente. Após sua purificação, as condições ótimas de pH e temperatura para a atividade catalítica de cada enzima foram determinadas, bem como sua termo estabilidade estrutural em diferentes tampões e pHs. Através de modelagem preditiva, as estruturas tridimensionais de ambas as enzimas foi avaliada. Por fim, a atividade dessas glucanases na inibição da formação e degradação de biofilmes pré-formados de Streptococcus mutans crescidos em meio de cultura contendo 0,1 ou 1% de sacarose foi avaliada em termos de quantificação de biomassa total, polissacarídeos insolúveis, microscopia confocal e produto de hidrólise. Os resultados sugerem que tanto a PmGH87 quanto a CoGH66 são capazes de inibir a formação dos biofilmes crescidos em condição limitada ou abundante de sacarose. Entretanto, a combinação de ambas não resulta em efeito sinérgico. Os dados de degradação de biofilmes pré-formados apontam um efeito tempo-dependente para o tratamento com cada enzima sozinha. Além disso, observou-se um efeito sinérgico e dose-dependente proveniente do tratamento enzimático combinado das glucanases, onde a maior degradação observada foi de 95,5% em 30 minutos de tratamento para o biofilme crescido em baixa concentração de sacarose, e 93,8% após 2 horas de tratamento para o biofilme crescido em condição abundante do açúcar, cujos graus de sinergismo calculados foram de 5,54 e 3,18, respectivamente. Em conclusão, ambas as glucanases estudadas neste trabalho são efetivas na inibição da formação e na degradação de biofilmes previamente formados de S. mutans. Esses resultados são essenciais para o desenvolvimento de aplicações biotecnológicas e biomédicas que visam o controle de biofilmes bacterianos.

Dental biofilms characterize an oral health problem as they can lead to the development of caries and other very costly oral diseases. Its main components are insoluble extracellular polysaccharides. Such constituents are directly related to the cariogenicity of the bacterial communities and are formed by (1 → 3)- α e (1 → 6)-α glucosidic bonds. Considering the chemical structure of these molecules and their importance for the dental biofilm, the use of exogenous enzymes which are capable of cleaving these glucosidic linkages, called glucanases, might be a promising strategy for biofilms control. In the present work, a (1 → 3)-α (PmGH87) and a (1 → 6)-α (CoGH66) glucanase from Prevotella melaninogenica and Capnocytophaga ochraceae, respectively, were cloned and heterogously expressed. After their purification, optimal conditions of pH and temperature for the catalytic activity of each enzyme were established, as well as their structural thermal stability in different buffers and pHs. Through predictive modeling, the tridimensional structures of both enzymes were assessed. Finally, the activity of these glucanases in inhibiting the formation and in the degradation of previously formed Streptococcus mutans biofilms grown in culture media containing 0,1 or 1% of sucrose were evaluated in terms of total biomass, insoluble polysaccharides content, and product of hydrolysis quantification, as well as confocal microscopy. Results suggest that both PmGH87 and CoGH66 are capable of inhibiting biofilm formation grown in limited or abundant sucrose conditions. However, the combinatory effect of the two enzymes does not result in a synergistic effect. Data of degradation of preformed biofilms experiments indicate a time-dependent effect for the treatment with each enzyme alone. In addition, it was observed a synergistic and dose-dependent effect from the combined enzymatic treatment of glucanases, where the highest observed degradation was 95,5% in 30 minutes of treatment for the biofilm grown in low sucrose concentration, and 93,8% after 2 hours treatment for the biofilm grown in sugar-abundant condition, whose calculated degree of synergisms were 5,54 and 3,18, respectively. In conclusion, both glucanases herein studied are effective in inhibiting the formation and in degrading previously formed S. mutans biofilms. These results are essential for the development of biotechnological and biomedical applications engaged in controlling bacterial biofilms.
