A malária é uma doença que acomete principalmente regiões tropicais. Essas regiões também são conhecidas pelo baixo desenvolvimento econômico e, consequentemente, pelas dificuldades de tratamento contra doenças. Esse cenário se torna ainda mais grave com os casos emergentes de parasitos resistentes às terapias atuais. Neste contexto, esse projeto teve como objetivo a descoberta de novos híbridos de derivados de artemisinina com marinoquinolinas como inibidores do parasito causador da forma mais grave da malária, o Plasmodium falciparum. Assim, em parceria com o Prof. Dr. Carlos Roque Duarte Correia (UNICAMP), foram sintetizados 4 híbridos (HB01, HB02, HB03 e HB04) para a caracterização das propriedades inibitórias in vitro contra o parasito. Todos os compostos apresentaram atividade antiplasmodial contra a cepa 3D7 (sensível a cloroquina) na faixa do baixo nanomolar (IC50s entre 3,5 nM a 10 nM). Além disso, os híbridos não apresentaram efeito citotóxico em células de hepatocarcinoma (HepG2, IC50 > 50 μM), determinando valores de índice de seletividade (IS) superiores a 3.000. O híbrido HB01, representativo da série, teve sua atividade inibitória avaliada frente a um painel de cepas resistentes do parasito (e.g., K1, TM90C6B e 3D7R_848) mostrando atividade promissora em todas as cepas avaliadas (IC50s entre 6 nM e 7 nM). Para verificar a eficácia dos híbridos quando comparados aos seus compostos de origem, foi realizado um ensaio de combinação. Nesse ensaio, verificou-se que os híbridos HB02 e HB04 foram mais potentes que a combinação de seus compostos de origem testados em concentrações equimolares. Os quatro híbridos foram avaliados quanto a sua atividade antiplasmodial na cepa de P. falciparum IPC 4912, que apresenta resistência parcial a derivados de artemisinina quando utilizado o ensaio de sobrevivência de anéis (RSA). Os valores de IC50 obtidos neste ensaio (e. g., 2,9 nM a 4,23 nM) foram semelhantes aos derivados de artemisinina. Diante disso, ensaios RSA e de recrudescência (RA) foram realizados e comparados com o controle dihidroartemisinina (DHA). Esses ensaios indicaram que o híbrido HB04 não apresentou anéis viáveis após 3 dias de experimento e não permitiu o recrudescimento dos parasitos até o final do experimento, 7 dias. Análise comparativa das propriedades antiplasmodiais dos híbridos avaliadas nesta dissertação indicou que o híbrido HB04 é uma molécula potente contra cepas sensível e resistentes do parasito, com baixa atividade citotóxica, capaz de impedir a sobrevivência das formas de anéis e a recrudescência do parasito. Portanto, HB04 é uma molécula bioativa que apresenta um conjunto de atividades antiplasmodiais promissora para o desenvolvimento de novos fármacos para a malária.

Malaria is a disease that mainly affects tropical regions. These regions are known for their low economic development and, consequently, for the difficulties of treating diseases. This scenario becomes even more serious with the emerging cases of parasites resistant to current therapies. Thus, this project aimed to discover new hybrids of artemisinin derivatives with marinoquinolines as inhibitors of the parasite that causes the most severe form of malaria, Plasmodium falciparum. Thus, in collaboration with Prof. Dr. Carlos Roque Duarte Correia (UNICAMP) four hybrids (HB01, HB02, HB03 and HB04) were synthesized and assessed against the parasite. All compounds showed potent antiplasmodial activity against the 3D7 strain (chloroquine-sensitive) with IC50 values between 3.5 nM to 10 nM. Furthermore, the hybrids showed no cytotoxic effect against hepatocarcinoma cells (HepG2, IC50 > 50 μM), determining selectivity index (SI) values greater than 3,000. The inhibitory activity of the hybrid HB01, representative of the series, was assessed against a panel of resistant strains of the parasite (e.g., K1, TM90C6B, and 3D7R_848), showing potent inhibition against all strains (IC50s between 6 nM and 7 nM). To verify the effectiveness of the hybrids when compared to their parent compounds, a combination assays were performed. In these assays, two hybrids (e.g., HB02 and HB04) showed enhanced inhibitory potency compared to the combination of their parent compounds when tested at equimolar concentrations. Moreover, the inhibitory activity of the hybrids was evaluated against the IPC 4912 strain of P. falciparum, which shows partial resistance to artemisinin derivatives in the ring survival assay (RSA). The IC50 values obtained ranged from 2.9 nM to 4.23 nM. The assessed IC50 values (e.g., 2.9 nM to 4.23 nM) were similar to artemisinin derivatives. Therefore, the RSA and recrudescence (RA) assays were performed, and the results compared to the control dihydroartemisinin (DHA). These assays indicated that the HB04 hybrid showed no viable rings after 3 days of experiment, in addition, the hybrid prevented the recrudescence of the parasites until the end of the experiment at day 7. The comparative analysis of the antiplasmodial properties of the hybrids assessed in this dissertation indicated that the HB04 hybrid is a potent molecule against sensitive and resistant strains of the parasite, with low cytotoxic activity, capable of preventing the survival of ring forms and the recrudescence of the parasite. Therefore, HB04 is a bioactive molecule showing promising antiplasmodial activities for the development of new drugs for malaria.
