A demanda global por energia tem acompanhado o desenvolvimento de novas indústrias, o que exige um constante aprimoramento e busca por novas fontes de energia. Com a conscientização da população a respeito da necessidade de fontes de energia renovável se abriu uma expectativa otimista para o uso da biomassa como fonte de energia. Uma variedade de microrganismos, como bactérias e fungos produzem enzimas que podem ser usadas para a despolimerização de biomassa celulósica em monômeros de glicose. Dentro deste contexto, estudamos uma celulase (XCC3535) de Xanthomonas campestris. Esta proteína é um híbrido natural com dois domínios (um domínio glicosilhidrolase e um domínio semelhante à expansina) que podem possuir funções complementares. Acredita-se que expansinas e proteínas semelhantes a expansinas possam ajudar na degradação da celulose cristalina, participando na ligação ao substrato, seguida pela formação de complexos enzimáticos ou, possivelmente, associadas à superfície para degradação. Este trabalho foi iniciado com análises de bioinformática da sequência para a proteína Glicosilhidrolase permitindo três construções para clonagem : uma completa contendo ambos os domínios (catalítico e semelhante a Expansina), outro com apenas o domínio catalítico GH5, e uma terceiro com apenas o domínio semelhante a Expansina. Obtemos por expressão recombinante em Escherichia coli Rosetta proteína das três construções. A amostra protéica pura foi utilizada para estudos por espalhamento dinâmico de luz (DLS), espalhamento de raios-X a baixo ângulo (SAXS), dicroísmo circular (CD) e cristalização para uso aplicando difração de raios-X. Assim, nosso principal objetivo neste estudo foi a descrição da Glicosilhidrolase XCC3535, permitindo assim a geração de conhecimento para o entendimento da função desta proteína e de seus domínios.

Global demand for energy has grown with the development of new industries that requires constant improvement and search for new sources of energy. Nowadays, the growing awareness of the population about the need for renewable energy have opened an optimistic expectation for the use of biomass as energy source. A variety of microorganisms like bacteria and fungi produce enzymes that can be used for the depolymerization of cellulosic biomass into glucose monomers. Within this context, we study one cellulase (XCC3535) from Xanthomonas campestris organism. This protein is a natural hybrid with two domains (a glicosyl hidrolase domain and an expansin-like domain) which seem to have complementary functions. It is believed that expansins help in the degradation of crystalline cellulose by participating in substrate binding, followed by formation of enzyme complexes or possibly attaching to cell surface. This work was initiated using bioinformatics analysis of the protein sequence and three constructs were made: a full length containing both domains (catalytic and expansin), another with only the catalytic domain, and a third one with only the expansin-like domain. We were able to overexpress these recombinant proteins in Escherichia coli Rosetta strain. The pure protein sample was used for studied by Dynamic Light Scattering (DLS), Small angle X-ray scattering, Circular Dichroism (CD) and Crystallization for X-ray diffraction. Thus, our main objective in this study was structurally describe these proteins, thereby allowing the generation of knowledge for understanding function of these proteins.
