A 2,3-butanodiona é utilizada em diferentes indústrias como flavorizante, conservante, componentes de fragrâncias etc., apesar disso, a exposição prolongada está associada a doenças e alterações proteicas. Diante deste fato, este estudo tem como principal objetivo a determinação proteômica em amostras de cérebros de ratos da linhagem Wistar (Rattus norvegicus) expostos cronicamente a 2,3-butanodiona. O conjunto amostral foi composto por 12 ratos (2 machos e 2 fêmeas), separados em três grupos, o primeiro grupo, controle, recebeu apenas o veículo, os outros dois grupos receberam 2,3butanodiona via intragástrica, nas dosagens de 46,6 µg kg-1 dia-1 e 99,1 µg kg-1 dia-1 durante 90 dias. Após o tempo de tratamento, os animais foram mortos por aprofundamento em anestesia e as amostras foram coletadas. As análises utilizaram técnicas cromatográficas e de espectrometria de massas (Nano-LC e MALDI-IMS) além do tratamento quimiométricos dos dados obtidos pelos equipamentos. Os dados coletados no Nano-LC foram analisados usando o software Skyline, que identificou 9296 proteínas, destas 2500 foram uteis para o tratamento dos dados quimiométricos que foi feito com o utilizando o MetaboAnalyst 5.0 e comparou-se os grupos com e sem distinção de sexo. Os testes realizados sem distinguir o sexo dos animais já mostrou uma separação dos grupos mostrando que a presença do aditivo pode alterar o perfil proteico do cérebro. Quando separado os animais machos e fêmeas também foi possível observar a separação dos grupos demonstrando que ambos os sexos são afetados pela exposição a 2,3-butanodiona. Em seguida as proteínas com maior influência foram listadas e foram correlacionadas com as doenças que sua alteração pode causar. O imageamento espectrométrico apontou diferenças na intensidade e na distribuição dos íons quando comparados os tecidos dos grupos tratados e do grupo controle.

Because of this fact, the main objective of this study is the proteomic determination in brain samples from Wistar rats (Rattus norvegicus) chronically exposed to 2,3-butanedione. The sample-set consisted of 12 rats (2 males and two females), separated into three groups, the first group, control, received only the vehicle, the other two groups received 2,3 - butanedione intragastric, at dosages of 46, 6 µg kg-1 day-1 and 99.1 µg kg-1 day-1 for 90 days. After the treatment, the animals were killed by deep anesthesia, and samples were collected. The analyzes used chromatographic and mass spectrometry techniques (Nano-LC and MALDI-IMS) in addition to the chemometric treatment of the data obtained by the equipment. The data collected in the Nano-LC were analyzed using the Skyline software, which identified 9296 proteins, of these 2500 was helpful in the treatment of the chemometric data that was carried out using the MetaboAnalyst 5.0 and the groups with and without distinction of sex were compared. The tests carried out without distinguishing the sex of the animals have already shown a separation of the groups indicating that the presence of the additive can change the protein profile of the brain. When the male and female animals were separated, it was also possible to observe the separation of the groups, demonstrating that both sexes are affected by exposure to 2,3-butanedione. Then the proteins with the most significant influence were listed and correlated with the diseases that their alteration can cause. The spectrometric imaging also showed differences in the intensity and distribution of ions when comparing the tissues of the treated groups and the control group.
