Lipases (glicerol éster hidrolases, E.C. 3.1.1.3) compreendem um grupo de enzimas hidrolíticas. Sabe-se que a lipase age na interface óleo-água e, por essa razão, a área interfacial livre total é um grande requisito de aumentar o rendimento de diferentes reações. Em termos comerciais, as enzimas imobilizadas são preferidas em relação às enzimas livres por apresentarem maior estabilidade, especificidade, atividade e por possibilitar a reutilização do biocatalisador. Dentre os métodos para imobilização, estão a encapsulação, podendo ser na matriz ou na membrana ou ligação, por adsorção ou ligação covalente. Um material utilizado como matriz no processo de imobilização é a casca de ovo de galinha. Essa matriz é considerada um resíduo inerte de menor custo e de fácil acesso, além de agregar valor quando utilizada no processo. Um mecanismo interessante que pode ser usado para acelerar as taxas iniciais de muitas reações químicas é a utilização do ultrassom. Essa técnica tem mostrado resultados favoráveis na melhora do desempenho de diferentes enzimas, como também, é considerada uma "química verde", por causa da sua eficiência, necessidade de pouca instrumentação, redução do tempo do processo versus técnicas convencionais tornando o processo economicamente viável. Neste contexto, esta dissertação teve como objetivo produzir lipase de Geotrichum candidum por fermentação utilizando subprodutos da indústria de alimentos (torta de milho e água de maceração de milho), realizar a imobilização da lipase em casca de ovo em pó e, por fim, comparar a atividade da lipase livre e imobilizada para verificar o rendimento da esterificação enzimática com aplicação da técnica de ultrassom. LL é definida como lipase livre liofilizada e LIC é a lipase imobilizada em casca de ovo em pó. Para verificar a porcentagem de adsorção de lipase em casca de ovo, foi feito um delineamento fatorial 2³ com as variáveis independentes pH da solução tampão fosfato (6 e 8), concentração da enzima (200 e 400 U/g) e temperatura (27 °C e 37°C). LL e LIC foram conduzidas em banho de ultrassom, 40 kHz, 132 W a 36° C, de 0 a 55 min, fez o acompanhamento da atividade enzimática e foi obtida a melhor condição para reação de esterificação: proporção molar 1:3 de ácido oléico e álcool metílico, 7,5 U/g de lipase liofilizada, 50° C no agitador orbital por 1h, 200 rpm. LL apresentou 1875 U/g de atividade e a melhor condição da LIC foi: pH 8, 400 U/g de lipase e 27°C resultando em 107% de adsorção. LL e LIC aumentaram sua atividade enzimática em 5 minutos de banho de ultrassom obtendo um ganho de 14,29% e 56,47% em relação ao início. Os resultados foram promissores demonstrando eficiência na imobilização da lipase em casca de ovo em pó para aplicação no banho de ultrassom, como também, na reação de esterificação.

Lipases (glycerol ester hydrolases, E.C. 3.1.1.3) comprise a group of hydrolytic enzymes. It is known that lipase acts at the oil-water interface and, for this reason, the total free interfacial area is a major requirement to increase the yield of different reactions. In commercial terms, immobilized enzymes are preferred over free enzymes because they have greater stability, specificity, activity and allow the reuse of the biocatalyst. Among the methods for immobilization, are encapsulation, which can be in the matrix or in the membrane or bond, by adsorption or covalent bond. A material used as a matrix in the immobilization process is chicken egg shell. This matrix is considered to be an inert residue of lower cost and easy access, in addition to adding value when used in the process. An interesting mechanism that can be used to accelerate the initial rates of many chemical reactions is the use of ultrasound. This technique has shown favorable results in improving the performance of different enzymes, as well as, it is considered a "green chemistry", because of its efficiency, the need for little instrumentation, reduction of the process time versus conventional techniques making the process economically viable. In this context, this dissertation aimed to produce Geotrichum candidum lipase by fermentation using by-products from the food industry (corn steep liquor and corn cake), to perform the immobilization of the lipase in powdered eggshell and, finally, to compare the activity of free and immobilized lipase to verify the performance of enzymatic esterification with the application of the ultrasound technique. LL is defined as lyophilized free lipase and LIC is the lipase immobilized in powdered eggshell. To verify the percentage of lipase adsorption in eggshell, a factorial design 23 was carried out with the independent variables pH of the phosphate buffer solution (6 and 8), enzyme concentration (200 and 400 U/g) and temperature (27 °C and 37°C). LL and LIC were carried out in an ultrasound bath, 40 kHz, 132 W at 36° C, from 0 to 55 min, followed the enzymatic activity and the best condition for the esterification reaction was obtained: 1: 3 molar ratio of oleic acid and methyl alcohol, 7.5 U/g of lyophilized lipase, 50° C in the orbital shaker for 1h, 200 rpm. LL showed 1875 U/g of activity and the best condition of the LIC was: pH 8, 400 U/g of lipase and 27°C resulting in 107% adsorption. LL and LIC increased their enzyme activity in 5 minutes of ultrasound bath obtaining a gain of 14.29% and 56.47% in relation to the beginning. The results were promising, showing efficiency in the immobilization of lipase in eggshell powder for application in the ultrasound bath, as well as in the esterification reaction.
