A depressão é um grande problema global, que foi agravada com a recente pandemia de COVID 19, o consumo de antidepressivos por parte da população aumentou e consequentemente sua detecção no ambiente. Os antidepressivos são fármacos considerados contaminantes ambientais e podem provocar mudanças comportamentais que afetam a dinâmica das populações mesmo em baixas concentrações. A ecotoxicologia é um ramo da toxicologia que se preocupa com o estudo dos efeitos tóxicos, causados por poluentes naturais e sintéticos, aos constituintes dos ecossistemas. Daphnia magna é um microcrustáceo amplamente utilizado em ensaios ecotoxicológicos agudo e crônico. O objetivo deste estudo foi avaliar a ocorrência de antidepressivos residuais como a fluoxetina (FLU), citalopram (CIT), venlafaxina (VEN) e sertralina (SER) em quatro pontos Rio Piracicaba, SP e realizar teste de toxicidade aguda e crônica em Daphnia magna. Para as análises foi utilizada a metodologia de extração em fase sólida (SPE) e um cromatógrafo líquido acoplado a um espectrômetro de massas com fonte de ionização electrospray (HPLC MS/MS). Os parâmetros utilizados para validação seguiram as recomendações da Anvisa. Os ensaios ecotoxicológicos agudos e crônicos com Daphnia magna seguiram as recomendações da Associação Brasileira de Normas Técnicas e Organisation for Economic Co-operation and Development. Com a realização do teste agudo, determinou-se a CE50 de cada substância, e foi possível realizar uma classificação quanto a toxicidade. Calculou-se o quociente de risco agudo para cada substância. Um teste agudo e crônico foi realizado com a maior concentração ambiental determinada, utilizando as substâncias individuais e suas misturas. No teste crônico foram avaliadas a reprodução, sobrevivência, tamanho dorsal e ventral. O método cromatográfico mostrou-se exato e preciso. Todos os antidepressivos estudados foram detectados nas amostras coletadas, na faixa de concentração de 1,0 a 4,48 ng L-1. A classificação efetuada mostrou que a FLU e o CIT são moderadamente tóxicos, a VEN pouco tóxica e a SER foi considerada muito tóxica. O quociente de risco para os antidepressivos estudados não apresentam risco ambiental agudo para Daphnia magna. Os testes agudos com a mistura dos antidepressivos nas concentrações ambientais não apresentaram organismos mortos ou imóveis. Os testes crônicos demonstraram efeitos na reprodução e tamanho dorsal da Daphnia magna, tanto os realizados com a substância individual quanto suas misturas. O tamanho ventral organismo não foi afetado pela exposição individual das substâncias, porém a mistura dos fármacos CIT.FLU.SER e VEN.CIT.FLU.SER propiciou os efeitos que mais se afastaram do controle. A variável sobrevivência não atingiu as pressuposições para avaliação estatística. Contudo, espera-se que esta pesquisa possa contribuir com o despertar do interesse das autoridades ambientais, para que haja um aprofundamento nos estudos ecotoxicológicos e melhor caracterização dos riscos associados à introdução contínua dessas substâncias no ambiente

Depression is a major global problem which was aggravated by the recent pandemic of COVID 19. The consumption of antidepressants by the population increased and, therefore, its detection in the environment. Antidepressants are drugs considered to be environmental contaminants and can cause behavioral changes that affect population dynamics even at low concentrations. Ecotoxicology is a branch of toxicology that is concerned with the study of toxic effects, caused by natural and synthetic pollutants, to the constituents of ecosystems. Daphnia magna is a microcrustacean widely used in acute and chronic ecotoxicological tests. The objective of this study was to evaluate the occurrence of residual antidepressants such as fluoxetine (FLU), citalopram (CIT), venlafaxine (VEN) and sertraline (SER) in four different spots of the Piracicaba River, in order to perform an acute and chronic toxicity test on Daphnia magna. The samples were analyzed through solid phase extraction methodology (SPE) and a liquid chromatograph coupled to a mass spectrometer with electrospray ionization source (HPLC MS / MS). The parameters used for validation followed Anvisa's recommendations. The acute and chronic ecotoxicological tests with Daphnia magna followed the recommendations of the Brazilian Association of Technical Standards and Organization for Economic Co-operation and Development. The performance of the acute test allows to determine the EC50 of each substance and it was possible to carry out a classification by toxicity. We also calculated the acute risk quotient for each substance. And performed an acute and chronic test with the highest environmental concentration determined, using the individual substances and their mixtures. Reproduction, survival, dorsal and ventral size were evaluated in the chronic test. The chromatographic method proved to be accurate and precise. All antidepressants studied were detected in the collected samples, in the concentration range of 1.0 to 4.48 ng L-1. The proposed classification showed FLU and CIT as moderately toxic, VEN as not very toxic and SER as very toxic. The risk quotient for the studied antidepressants does not present an acute environmental risk for Daphnia magna. Acute tests with the mixture of antidepressants at environmental concentrations did not show dead or immobile organisms. The chronic tests demonstrated effects on the reproduction and dorsal size of Daphnia magna, both performed with the individual substance and its mixtures. The ventral organism size was not affected by the individual exposure to the substances; however, the mixture of the drugs CIT.FLU.SER and VEN.CIT.FLU.SER propitiated the effects that most moved away from the control. The survival variable did not reach the assumptions for statistical evaluation. However, we hope that this research can contribute to the awakening of environmental authorities interest, so that there is a deepening in ecotoxicological studies and a better characterization of the risks associated with the continuous increase of these substances in the environment
