Em Angiospermas, o pistilo, contido na flor, é um órgão de extrema importância na reprodução vegetal, sendo que analisar a função dos genes expressos nos tecidos especializados que o compõem, é fundamental para um melhor entendimento do processo reprodutivo vegetal. Utilizando a técnica de screening diferencial de uma biblioteca de cDNA de estigma/estilete de Nicotiana tabacum, foi identificado um clone de cDNA (PA3), com alta similaridade a metiltransferases (SAMT, BAMT, e JAMT). O gene correspondente ao cDNA (PA3) foi denominado NtPMT (Nicotiana tabacum Pistil Methyltransferase). As metiltransferases são enzimas envolvidas em processos como desenvolvimento vegetal, respostas de defesa, sinalização química, polinização, entre outros. Dois clones de cDNAs, (46B11 e 134B02), codificando proteínas com alta similaridade a metiltransferases, foram isolados de um banco de ESTs (TOBEST), e utilizados para experimentos de expressão heteróloga. Para tanto, foram construídos dois plasmídeos recombinantes, contendo uma "tag" de histidinas em fusão N-terminal com as regiões codificadoras das proteínas, sob controle de um promotor forte e regulável. Os vetores de expressão bacterianos foram utilizados para transformar diferentes linhagens de Escherichia coli [BL21 (DE3), BL21(DE3) Códon Plus-RP e BL21 (DE3) Rosetta], de forma a analisar os níveis de expressão em diferentes temperaturas de cultivo (37°C, 30°C e 20°C) e diferentes tempos de indução (1, 2, 3, 4, 5 e 20hs). O vetor de expressão pEXP17¬46B11 foi utilizado para transformar as três linhagens de E. coli, enquanto o vetor de expressão pEXP17-134B02 foi introduzido apenas em BL21(DE3) Rosetta. Em BL21 (DE3) a proteína esperada (22,7kDa), correspondente ao clone pEXP17-46B11, foi produzida apenas em 37°C, apresentando um bom nível de expressão já nas primeiras três horas de indução. Por outro lado, as cepas BL21 (DE3)Códon Plus-RP e BL21 (DE3) Rosetta foram capazes de produzir boa quantidade da proteína recombinante, em todas as condições testadas. Entretanto, a proteína de fusão (22,7kDa) pôde ser observada apenas na fração celular insolúvel, na maioria das condições testadas, provavelmente na forma de corpos de inclusão. Contudo, ao se utilizar 20h de indução, na temperatura de 20°C, foi possível detectar uma pequena quantidade da proteína recombinante pEXP17-46B11 na fração celular solúvel, resultado confirmado por análises de Western blot. A linhagem BL21(DE3) Rosetta também apresentou bons níveis de expressão da proteína recombinante pEXP17-134B02, em todas as condições testadas. Entretanto, a proteína, com massa esperada (13,9kDa), foi observada apenas na fração celular insolúvel, provavelmente na forma de corpos de inclusão. Apesar de a proteína pEXP17-46B11 estar insolúvel, os bons níveis de expressão obtidos tornaram possível a utilização da mesma para a produção de anticorpos policlonais. Para tanto, bandas da proteína recombinante, induzidas em BL21(DE3) Rosetta, por três horas, à 37ºC, foram cortadas do gel de poliacrilamida, e utilizadas para a imunização de camundongos. Dois fragmentos do gene NtPMT foram amplificados via PCR, a partir do DNA genômico de N. tabacum e de suas prováveis espécies parentais, Nicotiana sylvestris e Nicotiana tomentosiformis. Após clonagem e sequenciamento, os fragmentos obtidos foram utilizados para análises comparativas de seqüências, visando um melhor entendimento da história evolutiva desse gene, nas três espécies de Nicotiana. Os resultados obtidos corroboraram com uma das hipóteses de origem evolutiva, segundo a qual, a espécie N. tabacum teria surgido a partir de um cruzamento entre um ancestral de N. sylvestris e um ancestral de N. tomentosiformis.

In Angiosperms, the pistil of the flower is an organ of extreme importance in plant reproduction and to analyze the function of the genes expressed in the specialized tissues of the pistil is very important for a better understanding of the reproductive process. Through the differential screening of a Nicotiana tabacum stigma/style cDNA library, a cDNA clone (PA3), encoding a protein with high similarity to methyltransferases (SAMT, BAMT and JAMT), has been identified. The gene corresponding to the cDNA (PA3) has been designated NtPMT (Nicotiana tabacum ? Pistil Methyltransferase). The methyltransferases are enzymes involved in processes such as: plant development, defense responses, chemical signaling, pollination, among others. Two cDNA clones (46B11 and 134B02), encoding proteins with high similarity to methyltransferases have been isolated from the TOBEST cDNA library and have been used in heterologous expression experiments. For this purpose, two recombinant plasmids have been constructed, containing a histidine tag in N-terminal fusion with the regions encoding the proteins, under the control of a strong and regulated promoter. The bacterial expression vectors were used for the transformation of different Escherichia coli strains [BL21 (DE3), BL21(DE3) Codon Plus-RP and BL21 (DE3) Rosetta], in order to analyze the expression levels in different growth temperatures (37°C, 30°C and 20°C) and different induction periods (1, 2, 3, 4, 5 and 20 hours). The expression vector pEXP17¬46B11 was used to transform the three strains of E. coli, and the expression vector pEXP17-134B02 has been introduced only in BL21(DE3) Rosetta. In BL21 (DE3), the expected protein (22.7kDa), corresponding to the pEXP17-46B11 clone, has been produced only at 37°C, showing a good expression level already at the first three hours of induction. On the other hand, the strains BL21 (DE3) Codon Plus-RP and BL21 (DE3) Rosetta were capable of producing good quantity of the recombinant protein in all conditions tested. However, the fusion protein (22.7kDa) could be observed only at the insoluble cellular fraction, in the majority of conditions tested, probably as inclusion bodies. However, using 20 h of induction at the temperature of 20°C, it was possible to detect a small quantity of the pEXP17-46B11 recombinant protein in the soluble cellular fraction, a result that has been confirmed by Western blot analyzis. The strain BL21(DE3) Rosetta has also shown good expression levels of the pEXP17-134B02 recombinant protein in all conditions tested. However, the protein with the expected mass (13.9kDa) has been observed only in the insoluble cellular fraction, probably as inclusion bodies. Despite the fact the pEXP17-46B11 protein was insoluble, the good expression levels obtained made possible its use in the production of policlonal antibodies. For this purpose, bands of the recombinant protein obtained in BL21(DE3) Rosetta, induced for three hours at 37ºC, were cut from the poliacrylamide gel and used for the immunization of mouses. Two fragments of the NtPMT gene were amplified by PCR, from genomic DNA of N. tabacum and its putative parental species Nicotiana sylvestris e Nicotiana tomentosiformis. After cloning and sequencing, the obtained fragments were used for comparative sequence analysis, aiming a better understanding of the evolutionary history of this gene in the three Nicotiana species. The results corroborate the hypothesis of the N. tabacum evolutionary origin in which it was originated by the cross between an ancestral of N. sylvestris and an ancestral of N. tomentosiformis.
