A população idosa vem aumentando ao longo dos anos com consequente incremento das doenças ósseas. Nesse contexto social, uma das estratégias mais promissoras para o tratamento de lesões ósseas é a engenharia de tecidos. Na engenharia de tecido ósseo, os scaffolds devem mimetizar as propriedades físicas da matriz extracelular óssea para facilitar o recrutamento, adesão, proliferação e diferenciação das células. Nanomateriais diversas vezes são associados aos scaffolds para buscar melhorar a estrutura, bioatividade, estabilidade, adesão celular e/ou compatibilidade. O Carbon dots (CDs) são um nanomaterial de carbono fluorescente com o diâmetro abaixo de 10 nm. O objetivo desse trabalho foi desenvolver os scaffolds de quitosanagelatina/hidroxiapatita, assim como os Carbon dots desejados para aplicação em Engenharia de Tecidos ósseos. Foram desenvolvidos inicialmente dois tipos de CD diferentes, sendo que um deles apresentou melhor rendimento quântico de fluorescência e citotoxicidade. Os scaffolds com e sem CDs preparados apresentaram, como esperado, características importantes para a sua aplicação, como a porosidade e taxa de degradação. Quanto aos testes in vitro obteve-se uma taxa de adesão celular excelente, boa citotoxicidade e expressão de fosfatase alcalina. Os testes in vivo do scaffolds antes da impregnação comprovaram que o material desenvolvido apresente excelente capacidade regenerativa para aplicação em engenharia de tecidos ósseos. Conclui-se, portanto, o material proposto atende as propostas estabelecidas e com resultados promissores.

The elderly population has been increasing over the years with a consequent increase in bone diseases. In this social context, one of the most promising strategies for the treatment of bone injuries is tissue engineering. In bone tissue engineering, scaffolds must mimic the physical properties of bone extracellular matrix to facilitate cell recruitment, adhesion, proliferation and differentiation. Nanomaterials are often associated with scaffolds to improve their structure, bioactivity, stability, cell adhesion and/or compatibility. Carbon dots (CDs) are a fluorescent carbon nanomaterial with a diameter below 10 nm. The objective of this work was to develop the chitosan-gelatin/hydroxyapatite scaffolds, as well as the desired Carbon dots for application in Bone Tissue Engineering. Two different types of CD were initially developed, one of which presented better quantum yield of fluorescence and cytotoxicity. The scaffolds with and without prepared CDs presented, as expected, important characteristics for their application, such as porosity and degradation rate. As for the in vitro tests, an excellent cell adhesion rate, good cytotoxicity and alkaline phosphatase expression were obtained. The in vivo tests of the scaffolds before impregnation proved that the material developed has excellent regenerative capacity for application in bone tissue engineering. It is concluded, therefore, that the proposed material meets the established proposals and with promising results.
