A lignocelulose representa a fonte de carbono renovável mais abundante da Terra, a partir da qual bioprodutos de alto valor agregado, como o etanol 2G, podem ser obtidos. Contudo, devido à elevada recalcitrância imposta pela complexa associação entre celulose, hemicelulose e lignina, grandes quantidades de enzimas do tipo CAZymes são necessárias para a conversão eficiente em açúcares fermentáveis. Os preços elevados dessas enzimas correspondem hoje a mais de 15% dos custos totais envolvidos no processamento da biomassa, sendo portanto um dos principais entraves para a implementação de biorrefinarias lignocelulósicas em larga escala e de forma economicamente viável. A bioprospecção de enzimas mais robustas para aplicações industriais pode contribuir para a otimização dos coquetéis enzimáticos, maximizando os rendimentos dos produtos e, simultaneamente, reduzindo os custos do processo. Nesse contexto, o objetivo do presente trabalho foi caracterizar as propriedades bioquímicas e cinéticas de três enzimas hidrolíticas de A. fumigatus Af293 - uma endoglucanase GH7 (Af-EGL7), uma celobiohidrolase GH6 (AfCel6A) e uma xilanase GH10 (Af-XYLA) - as quais foram identificadas pelo nosso grupo de pesquisa em secretoma e transcriptoma, quando o fungo foi cultivado na presença de bagaço de cana-de-açúcar explodido (SEB). Para tanto, o gene Af-egl7 foi clonado nos respectivos vetores pET-28a(+) e pPICZαA para expressão heteróloga em E. coli RosettaTM (DE3) pLysS e P. pastoris X-33. Os genes Afcel6A e Af-xylA, por sua vez, foram expressos somente em P. pastoris X-33. A endoglucanase Af-EGL7 produzida em dois sistemas de expressão diferentes apresentou propriedades enzimáticas bem distintas, principalmente em termos de estabilidade e eficiência catalítica. As condições ótimas de temperatura e pH foram as mesmas em ambos os casos - 55 ºC e pH 5,0 - assim como a tolerância a diferentes aditivos presentes no meio reacional. Embora substancialmente menos ativa, a Af-EGL7 produzida em bactéria foi capaz de atuar sinergicamente com o coquetel Celluclast®1.5L durante a degradação de SEB, e, assim, promover aumentos consideráveis nos percentuais de açúcares redutores liberados. Similarmente, a Af-EGL7 expressa em P. pastoris exibiu sinergismo com o coquetel comercial sobre esse e outros resíduos lignocelulósicos. A celobiohidrolase AfCel6A exibiu atividade máxima entre 55-60 ºC e pH 5,5-6,0. Essa enzima apresentou estabilidade considerável em diferentes condições de temperatura e pH, além de alta resistência à inibição por glicose em concentrações de até 250 mM. A AfCel6A atuou em sinergia tanto com o coquetel quanto com a Af-EGL7 na degradação de CM-Celulose e biomassas mais complexas, o que resultou em rendimentos de hidrólise muito superiores em comparação ao controle. Para a xilanase Af-XYLA, o melhor desempenho foi alcançado a 80 ºC e na faixa de pH de 5,0-7,0. Os parâmetros cinéticos da Af-XYLA sobre xilana foram muito superiores aos reportados na literatura para outras xilanases GH10 fúngicas. A Af-XYLA demonstrou elevada tolerância aos açúcares glicose e xilose em concentrações de até 250 mM, além de estabilidade considerável a diferentes temperaturas e pH. Similarmente ao observado para as duas celulases recombinantes, o alto grau de sinergismo entre a Af-XYLA e as celulases comerciais intensificou substancialmente a liberação de açúcares redutores a partir de bagaço de cana-de-açúcar pré-tratado. Juntos, os resultados obtidos apontaram essas três enzimas recombinantes como potenciais alvos para aplicações em biorrefinarias lignocelulósicas.

Lignocellulose represents the most abundant renewable carbon source on Earth, from which high value-added bioproducts, such as 2G ethanol, can be obtained. However, due to the high recalcitrance imposed by complex association between cellulose, hemicellulose and lignin, large amounts of CAZymes are needed for an efficient conversion into fermentable sugars. The high prices of these enzymes currently account for more than 15% of the total costs involved in biomass processing, being therefore one of the main obstacles to the economically feasible implementation of large-scale lignocellulosic biorefineries. Bioprospecting for more robust enzymes for industrial applications can contribute to the optimization of enzyme cocktails, maximizing product yields and, simultaneously, reducing process costs. In this context, the aim of the present work was to characterize the biochemical and kinetic properties of three hydrolytic enzymes from A. fumigatus Af293 - a GH7 endoglucanase (Af-EGL7), a GH6 cellobiohydrolase (AfCel6A) and a GH10 xylanase (Af-XYLA) - which were identified by our research group on secretome and transcriptome, when the fungus was cultivated in the presence of sugarcane exploded bagasse (SEB). For this purpose, Af-egl7 gene was cloned into the respective vectors pET-28a(+) and pPICZαA for heterologous expression in E. coli RosettaTM (DE3) pLysS and P. pastoris X-33. Afcel6A and Af-xylA genes were expressed only in P. pastoris X-33. Af-EGL7 endoglucanase produced in two diferente expression systems showed very distinct enzymatic properties, mainly in terms of stability and catalytic efficiency. The optimal conditions of temperature and pH were the same in both cases - 55 ºC and pH 5.0 - as well as the tolerance to different additives present in the reaction mixture. Although substantially less active, Af-EGL7 produced in bacteria was able to act synergistically with the Celluclast®1.5L cocktail during SEB degradation and thus promote considerable increases in the percentage of reducing sugars released. Similarly, Af-EGL7 expressed in P. pastoris exhibited synergism with the commercial cocktail on this and other lignocellulosic residues. AfCel6A cellobiohydrolase exhibited maximal activity between 55-60 ºC and pH 5.5-6.0. This enzyme showed considerable stability under different conditions of temperature and pH, in addition to high resistance to inhibition by glucose at concentrations up to 250 mM. AfCel6A acted synergistically with both cocktail and Af-EGL7 on the degradation of CM-Cellulose and more complex biomasses, which resulted in much higher hydrolysis yields compared to the control. For Af-XYLA xylanase, the best performance was achieved at 80 ºC and in the pH range 5.0-7.0. Af-XYLA kinetic parameters on xylan were much higher than those reported in the literature for other fungal GH10 xylanases. Af-XYLA showed high tolerance to glucose and xylose at concentrations up to 250 mM, in addition to considerable stability at different temperatures and pH. Similar to what was observed for the two recombinant cellulases, the high degree of synergism between Af-XYLA and commercial cellulases substantially boosted the release of reducing sugars from pretreated sugarcane bagasse. Together, the results obtained indicated these three recombinant enzymes as potential targets for applications in lignocellulosic biorefineries.
