Neste estudo, um biorreator de calicreína de pâncreas de porco (KLK), imobilizada em partículas magnéticas (MP), foi desenvolvido como uma ferramenta altamente ativa, estável e útil para a triagem de ligantes, representando uma abordagem inédita. Este biorreator, denominado IMER-KLK-MP, foi então aplicado no ensaio de pesca de ligantes, possibilitando a identificação de ligantes presentes em matrizes de origem natural e em uma coleção de compostos sintetizados como híbridos multifuncionais direcionados a múltiplos alvos, denominados PQM, detectados de forma off-flow por cromatografia líquida de alta eficiência acoplada à espectrometria de massas (LC-MS). O IMER-KLK-MP foi utilizado também em ensaios de atividade na busca por moduladores enzimáticos. Dentre os compostos testados, dois apresentaram resultados significativos de inibição, com valores acima de 40%, sendo estes os compostos PQM-277 e PQM-219. Além disso, exploramos a imobilização da KLK em capilares de sílica fundida, resultando na obtenção de KLK-ICERs para serem estudados de forma on-flow por LC-MS, também representando uma inovação. Essas abordagens propiciam avanços significativos ao proporcionar ferramentas que podem ampliar a busca por ligantes com potencial para o desenvolvimento de terapias direcionadas às KLKs.

In this study, a kallikrein porcine pancreas bioreactor (KLK), immobilized on magnetic particles (MP), was developed as a highly active, stable, and useful tool for ligand screening, representing a novel approach. This bioreactor, named IMER-KLK-MP, was then applied in a ligand fishing assay, enabling the identification of ligands present in matrices of natural origin and in a collection of synthesized compounds as multifunctional hybrids targeted to multiple receptors, designated as PQMs, detected off-flow by liquid chromatography-high-resolution mass spectrometry (LC-MS). The IMER-KLK-MP was also employed in activity assays in the search for enzymatic modulators. Among the tested compounds, two showed significant inhibition results, with values above 40%, namely, compounds PQM-277 and PQM-219. Additionally, we explored the immobilization of KLK in fused silica capillaries, resulting in the obtaining of KLK-ICERs to be studied on-flow by LC-MS, also representing an innovation. These approaches provide significant advances by offering tools that can broaden the search for ligands with potential for the development of therapies targeted to KLKs.
