Foi desenvolvido um sistema de microscopia especular para Banco de Olhos, para análise de córneas "in vitro", com várias magnificações: 20x, 30x, 63x, 136x e 204x. O sistema foi projetado num software comercial ZEMAX-XE, da ZEMAX Developmen Corporation. O sistema de iluminação projetado apresentou aproximadamente 60µm de aberração cromática, 0.8 unidades de onda (waves) de aberração esférica e nenhum astigmatismo. O sistema de magnificação apresentou para as magnificações de 20x, 30x, 63x, 136x e 205x, as respectivas aberrações cromáticas: ±250µm, ±200µm, ±200µm, ±300µm e ±25µm; as respectivas aberrações esféricas: -0.6 unidades de onda, -0.6 unidades de onda, -0.5 unidades de onda, 0.025 unidades de onda, e 0.001 unidades de onda, e nenhum astigmatismo para qualquer das magnificações. O sistema ótico montado em bancada, segundo o projeto desenvolvido apresentou as reais magnificações de 24x, 40x, 64x, 100x e 160x. As imagens obtidas de córneas humanas "in vitro" e densidades celulares comparadas a um sistema comercial, estão em concordância de 90% para as contagens interativas das células.

We have developed a specular microscopy system for Eye Banks for "in vitro" corneal analysis with several magnifications: 20x, 30x, 63x, 136x and 205x. The optical design of the system was projected using a commercial software ZEMAX-XE, from ZEMAX Developmen Corporation. The projected illumination system has presented approximately a chromatic aberration of 60µm, a spherical aberration of , 0.8 waves and no astigmatism. The magnification system has presented for the 20x, 30x, 63x, 136x and 205x magnifications, the respective chromatic aberrations: ±250µm, ±200µm, ±200µm, ±300µm and ±25µm; and the respective spherical aberrations: -0.6wave, -0.6wave, -0.5wave, 0.025wave, and 0.001wave and no astigmatism for any of the magnifications. The optical bench system, according to the projected design has presented the real magnifications of: 24x, 40x, 64x, 100x and 160x. The images of human "in vitro" corneas and the cellular density provided by the system are in good agreement with a commercial system (90% for the interactive of the cells).
