A transdução do sinal Wnt é responsável pela organização estrutural durante a embriogênese, participando da estimulação mitogênica, diferenciação celular e homeostase de tecidos adultos, incluindo o tecido ósseo. É bem estabelecido que a superfície de titânio com nanotopografia (Ti-Nano) favorece a diferenciação osteoblástica por modular as vias de sinalização das integrinas e proteínas ósseas morfogenéticas (BMPs). Porém, poucos estudos têm abordado a participação da via Wnt nos efeitos da topografia em nanoescala. Portanto, formulamos a hipótese de que a via de sinalização Wnt está, ao menos em parte, envolvida no maior potencial osteogênico da superfície de Ti-Nano. Para testar essa hipótese, utilizamos discos de Ti, que foram tratados com solução de H2SO4/H2O2 para produzir superfícies nanotopográficas, e discos sem tratamento (Ti-Usinado), sendo considerados como controle. Células da linhagem MC3T3-E1, subclone 14, foram cultivadas sobre ambos os discos para avaliar o efeito do Ti-Nano sobre a expressão de genes relacionados às vias de sinalização canônica Wnt/β-catenina e não canônica Wnt/Ca2+. Com base nos resultados do PCR em tempo real, os genes mais intensamente modulados pela superfície de Ti-Nano, Fzd4 relacionado à via Wnt/β-catenina e Fzd6 relacionado à via Wnt/Ca2+, foram selecionados e silenciados por CRISPR-dCas9-KRAB e CRISPR-Cas9, respectivamente. Então, foi investigado o efeito desses silenciamentos sobre o potencial osteogênico em células MC3T3-E1, cultivadas sobre Ti-Nano e Ti-Usinado. Os dados foram comparados pelo Teste t de Student ou pelo teste ANOVA one-way (p≤0,05). Os resultados deste estudo revelaram que: 1) o silenciamento do gene Fzd4 inibe a expressão do genótipo e fenótipo osteoblástico de células MC3T3-E1, coincidindo com a modulação negativa de alvos da via Wnt/β-catenina aos 5 dias sobre superfície de Ti-Nano; 2) o silenciamento de Fzd6 inibe a expressão do genótipo e fenótipo osteoblástico de células MC3T3-E1; no entanto, os alvos da via Wnt/Ca2+ não foram modulados, e observou-se expressão negativa da proteína β-catenina aos 3 dias de cultivo das células MC3T3-E1 sobre Ti-Nano. Estes resultados indicam que o maior potencial osteogênico da nanotopografia deve-se, ao menos em parte, à ativação da via de sinalização Wnt/β-catenina, uma vez que o silenciamento dos genes Fdz4 e Fzd6 reduziu intensamente a diferenciação osteoblástica de células MC3T3-E1 crescidas sobre Ti-Nano, comparado ao Ti-Usinado. Em conjunto, nossos resultados revelam um novo mecanismo que envolve a via Wnt/β-catenina para explicar o maior potencial osteogênico da superfície com nanotopografia aqui avaliada.

The Wnt signal transduction is crucial for the structural organization during embryogenesis, participating in mitogenic stimulation, cell differentiation and homeostasis of adult tissues, including bone. It is well established that the titanium surface with nanotopography (Ti-Nano) favors the osteoblastic differentiation by modulating the signaling pathways of integrins and bone morphogenetic proteins (BMPs). However, few studies have addressed the participation of the Wnt pathway on the effects of nanoscale topographies. Therefore, we hypothesized that the Wnt signaling pathway is, at least in part, involved in the higher osteogenic potential of Ti-Nano. To test this hypothesis, we used Ti discs, which were treated with H2SO4/H2O2 solution to produce nanotopographic surfaces, and discs without treatment (Ti-Machined), being considered as control. Then, MC3T3-E1 cells, subclone 14, were cultured on both surfaces to evaluate the effect of Ti-Nano on the gene expression related to the canonical Wnt/β-catenin and non-canonical Wnt/Ca2+ signaling pathways. Based on the real-time PCR results, the most intensely modulated genes by Ti-Nano, the Wnt/β-catenin pathway-related Fzd4 and Wnt/Ca2+ pathway-related Fzd6, were selected and silenced by CRISPR-dCas9-KRAB and CRISPR-Cas9, respectively. Then, we investigated the effect of the silencing on the osteogenic potential of Ti-Nano. Data were compared by Student's t-test or one-way ANOVA (p≤0.05). The results of this study revealed that: 1) Fzd4 gene silencing inhibits the osteoblast genotype and phenotype expression of MC3T3-E1 cells, corresponding to the negative modulation of Wnt/β-catenin targets in cells grown on Ti-Nano surface on day 5; 2) Fzd6 silencing inhibits the osteoblast genotype and phenotype expression of MC3T3-E1 cells; however, Wnt/Ca2+ pathway targets were not modulated, but negative expression of β-catenin protein was detected in cells grown on Ti-Nano on day 3. These results indicate that the higher osteogenic potential of Ti-Nano is, at least in part, due to the activation of the Wnt/β-catenin signaling pathway, since the silencing of the Fdz4 and Fzd6 gene significantly reduced the osteoblast differentiation of MC3T3-E1 cells grown on Ti-Nano, compared to Ti-Machined. Taken together, our results revealed a new mechanism involving the Wnt/β-catenin signaling pathway to explain the higher osteogenic potential of the evaluated nanotopography.
