O objetivo desse estudo foi avaliar o efeito do uso do extrato de Bromelina na resistência de união (RU) à dentina radicular de pinos de fibra de vidro. Inicialmente, determinou-se a concentração e tempo de aplicação mais eficientes da Bromelina. Assim, a suspensão do extrato de Bromelina nas concentrações de 0, 2%, 5% e 10%, foram aplicadas sobre dentina por 30 segundos ou 1 minuto. Os substratos foram caracterizados por meio da microscopia eletrônica de varredura (MEV). Para a análise de RU, foram selecionados dentes humanos unirradiculares hígidos que foram tratados, preparados para pino, e separados em 6 grupos, de acordo com o tratamento a que foram submetidos e tipo de cimento utilizado: cimento resinoso dual convencional (CDC); cimento resinoso dual autoadesivo (CDA); extrato de bromelina 5% (Br5%) + CDC; Br5% + CDA; extrato de bromelina 10% (Br10%) + CDC; Br10% + CDA. Metade das amostras de cada grupo foram submetidas ao ensaio de ciclagem termomecânica (CTM - Erios, 1.200.000 ciclos, carga axial de 133N e frequência de 2Hz) e a outra metade permaneceu 24h imersa em água destilada. As raízes dos dentes foram seccionadas transversalmente (2mm de espessura) e submetidas ao ensaio de push-out (Instrom, 2519-106, 0,5mm/min) nos terços cervical, médio e apical. Análises em MEV e Microscopia confocal foram utilizadas para caracterização da interface adesiva. Os dados de RU foram analisados com ANOVA de três fatores (p<0,05), Tukey. O teste qui-quadrado foi realizado para análise de prevalência dos padrões de fratura. Houve diferença (p<0,05) na RU para o fator cimento. Br10%+CDA apresentou menor RU (p<0,05) que quando utilizado CDC. Não houve diferença (p>0,05) entre terços quando utilizado o cimento CDA, com exceção do grupo tratado com Br10%, que apresentou menores valores de RU para o terço apical. Pinos cimentados com CDA resultaram em maior prevalência de fraturas adesivas à dentina. A mesma prevalência ocorreu após CTM nos grupos CDC e Br5%+CDC. Concluiu-se que o tratamento com Bromelina não interferiu na resistência de união entre pino e parede radicular em nenhuma concentração testada. O cimento autoadesivo apresentou menor resistência de união quando tratado com bromelina a 10%. Por fim, os pinos cimentados com cimento convencional apresentaram camada híbrida mais espessa e uniforme e quando cimentados com cimento autoadesivo houve formação de gaps entre cimento e dentina radicular.

Effect of bromelain on the bond strength of glass fiber posts using different resin cements

The aim of this study was to evaluate the effect of the use of Bromelain extract on the bond strength (BS) to the root dentin of glass fiber posts. First, it was selected the most efficient concentration and application time of Bromelain. Thus, a Bromelain extract suspension, of 0, 2%, 5% and 10%, were applied over the dentin for 30 seconds or 1 minute. The substrates were characterized using scanning electron microscopy (SEM). For the BS analysis, sound human single-rooted teeth were selected, treated, prepared for post, and separated in 6 groups, according to the treatment which it was submitted, and type of cement used: conventional dual resin cement (DC); dual self-adhesive resin cement (SAC); 5% bromelain extract (5%Br) + DC; 5%Br + SAC; 10% bromelain extract (10%Br) + DC; 10%Br + SAC. Half of the samples of each group were submitted to thermo-mechanical cycling (TMC - Erios, 1,200,000 cycles, axial load of 133N and frequency of 2Hz) and the other half was immersed in distilled water for 24h. The roots were transversely sectioned (2mm of thickness) and submitted to the push-out test (Instrom, 2519-106, 0.5mm/min) on the cervical, middle, and apical thirds. SEM analysis and confocal microscopy were used to characterize the hybrid layer. BS data were analyzed with 3-way ANOVA (p<0.05), Tukey's test. The chi-square test was performed to analyze the fracture pattern prevalence. There was difference (p<0.05) on the BS for the cement factor. 10%Br+SAC presented lower BS (p<0.05) than DC. There was no difference (p>0.05) between thirds when the SAC cement was used, with exception to the group treated with 10%Br, which presented lower BS values for the apical third. Posts cemented with SAC resulted in higher prevalence of adhesive to dentin fractures. The same prevalence occurred after TMC on the DC and 5%Br+DC groups. It was concluded that the Bromelain treatment did not interfere on the bond strength between post and root dentin in any concentration tested. The self-adhesive cement presented lower bond strength when treated with 10% bromelain. Lastly, posts cemented with conventional cement presented a hybrid layer thicker and more uniform, and when cemented with the self-adhesive cement there was gap formation between the cement and root dentin.
