Diferentes tratamentos de superfície são aplicados em implantes odontológicos de ligas de titânio a fim de modificar a superfície e alterar as respostas celulares. Tais modificações melhoram a osseointegração, entretanto, promovem topografia rugosa as quais estão sujeitas ao acúmulo de diferentes tipos de biofilme bacteriano, que diferem na quantidade e nas espécies colonizadoras. A obtenção de implantes odontológicos por meio da utilização da manufatura aditiva apresenta um implante com superfície topográfica e com caracterização química superficial diferente dos implantes disponíveis no mercado. Sabendo que a composição da superfície, estrutura química, molhabilidade, energia livre de superfície, forças eletrostáticas e rugosidade podem interferir no acúmulo de biofilme, o objetivo deste estudo foi avaliar três superfícies de discos de titânio obtidos por diferentes métodos, sendo 1 manufatura aditiva (DMA), 2 manufatura subtrativa com tratamento de superfície de hidroxiapatita (DUCT), 3 manufatura subtrativa sem tratamento (DUST), e compará-las quanto ao perfil microbiológico por meio de sequenciamento do gene 16S rRNA-based PCR. Oito corpos de prova com Ø 5,0 mm x 1 mm de altura foram obtidos por manufatura aditiva a partir do pó de Ti-6Al-4V, e 16 corpos de prova com Ø 8,0 mm x 3 mm de altura foram obtidos por manufatura subtrativa de barras cilíndricas de liga de Ti-6Al-4V. Destes 16 corpos de prova, 8 tiveram a superfície tratada com hidroxiapatita carbonatada (HAp) cultivada em fosfolipídio. Foram confeccionados aparelhos removíveis para alojar os discos a partir da moldagem da maxila de 8 pacientes. O aparelho foi utilizado por 96 horas, e o biofilme coletado a cada 24 horas. Foi realizada a extração do DNA genômico, o PCR 16S e o sequenciamento genético das amostras. A rugosidade foi avaliada por microscópio confocal a laser, e a composição química foi determinada por Espectroscopia de energia dispersiva de Raio-X (EDS). Para a análise estatística foi verificada a distribuição dos dados, e em seguida foram aplicados testes paramétricos, não-paramétricos e análise descritiva com nível de significância de 5% (α = 0,05). Quanto a rugosidade, foram observados diferentes níveis, sendo que a superfície DMA foi quem mostrou as maiores médias para todos os parâmetros avaliados (Ra, Rz e Sa). A análise da composição química por EDS mostrou predominância de Ti nas superfícies dos discos DMA e DUST, seguida pelos elementos Al e V. Nas superfícies do DUCT foi observada a predominância de O seguida de Ti, Ca, P e Mg. Com relação à quantificação microbiana total, foi observada diferença significante quando realizado o comparatório entre as três superfícies, sendo DUCT e DUST semelhantes na comparação múltipla (p= 0,80). Na análise de abundância relativa, foi observada semelhança na proporção dos gêneros (p= 0,99), sendo que Streptococcus (38,31%), Neisseria (10,43%) e Pasteurella (4,95%) foram os gêneros predominantes em uma contagem total sem considerar superfícies. Com relação á diversidade foi observada grande diversidade e sugere-se que não existem gêneros dominantes em nenhuma das superfícies investigadas.

Different surface treatments are applied on titanium alloy dental implants to modify the surface and cellular responses. Such modifications improve osseointegration and also promote rough topography, which is subject to the accumulation of different types of bacterial biofilm. These biofilms differ in the amount and colonizing species. Obtaining dental implants through additive manufacturing presents an implant with a topographic surface and a surface chemical characterization different from the implants available on the market. Knowing that the surface composition may interfere with biofilm accumulation, this study aimed to evaluate three surfaces of titanium discs obtained by different methods: 1 additive manufacturing (AMD), 2 subtractive manufacturing with hydroxyapatite surface treatment (SMDHT), 3 subtractive manufacturing without treatment (SMDWT), and compare them regarding the microbiological profile using 16S rRNA gene sequencing-based PCR. Eight specimens with Ø 5.0 mm x 1 mm height were obtained by additive manufacturing from Ti-6Al-4V powder. Sixteen specimens with Ø 8.0 mm x 3 mm height were obtained by subtractive manufacturing of cylindrical bars of Ti-6Al-4V alloy. Of these 16 specimens, 8 had the surface treated with carbonated hydroxyapatite (HAp) grown on phospholipid. Removable appliances were made to house the disks from the molding of the maxilla of 8 patients. The appliance was used for 96 hours, and the biofilm was collected every 24 hours. Genomic DNA extraction, 16S PCR, and gene sequencing of the samples were performed. The roughness was evaluated by laser confocal microscope, and the chemical composition was determined by Energy dispersive X-ray spectroscopy (EDS). For statistical analysis, data distribution was verified, and then parametric tests, non-parametric tests, and descriptive analysis were applied with a significance level of 5% (α = 0.05). As for roughness, different levels were observed, and the AMD surface was the one that showed the highest averages for all parameters evaluated (Ra, Rz, and Sa). The chemical composition analysis by EDS showed a predominance of Ti on the AMD and SMDWT disc surfaces, followed by the elements Al and V. On the DUCT surfaces a predominance of O was observed followed by Ti, Ca, P and Mg. Concerning total microbial quantification, a significant difference was observed when comparing the three surfaces, SMDHT and SMDWT being similar in the multiple comparison (p= 0.80). In the relative abundance analysis, similarity was observed in the proportion of genera (p= 0.99), and Streptococcus (38.31%), Neisseria (10.43%) and Pasteurella (4.95%) were the predominant genera in a total count without considering surfaces. Concerning diversity, a great diversity was observed, suggesting that there are no dominant genera on any of the surfaces investigated.
