O desenvolvimento de novas moléculas para inovação em diagnóstico de doenças crônicas, tais como câncer, aterosclerose e doenças neurodegenerativas, é foco de uma variedade de laboratórios de investigação ao redor do mundo. Atualmente, peptídeos, bem como suas diferentes abordagens de radiomarcação, têm tido um importante papel em testes clínicos em andamento, seja para diagnóstico ou tratamento, de vários tipos de cânceres. O objetivo deste trabalho é desenvolver a biomolécula sintética peptídeo-like [NOTA-CTHRSSVVC] radiomarcada com os isótopos [68Ga]gálio e [18F]flúor, para efeito de comparação. p-SCN-Bn-NOTA-CTHRSSVVC foi preparado pela reação de condensação utilizando os quelantes p-Bn-SCN-DOTA, ou p-Bn-SCN-NOTA, e o peptídeo cíclico CTHRSSVVC. O rendimento da reação, bem como os respectivos controles de qualidade dos produtos e suas purificações, foi realizado por cromatografia líquida de alta eficiência (CLAE) e os picos obtidos foram confirmados por espectrometria de massas. Em se tratando da radiomarcação, 80l do peptídeo conjugado (NOTA-CTHRSSVVC), obtido comercialmente, foi radiomarcado em 0,350 mL de solução de acetato de sódio 0,2M (pH 4,1), adicionando-se à solução 0,05 mL contendo 1,0 2,5 mCi (ou 37 92,5 MBq) de [68Ga]GaAcO ou [18F]AlF; as reações foram acompanhadas sob diferentes temperaturas e os controles de qualidade dos produtos radiomarcados foram avaliados por CLAE acoplado a um detector de radiação gama. A radiomarcação dos peptídeos conjugados [68Ga]Ga-NOTA-CTHRSSVVC e [18F]AlF-NOTA-CTHRSSVVC, purificados, foram analisados por CLAE, obtendo produtos com pureza radioquímica, de 97,8% (n=3) e 95,5% (n=3), respectivamente. Ambos os radiofármacos obtidos mantiveram-se estáveis quimicamente por 4 horas

The development of new molecules for innovation in the diagnosis of chronical diseases, such as cancer, atherosclerosis and neurodegenerative diseases, is the focus of a variety of researcher laboratories around the world. Currently, peptides, as well as their different approaches to radiolabeling, have played an important role in ongoing clinical tests, whether for diagnosis or treatment, of several types of cancers. The aim of this work is to develop the peptide-like synthetic biomolecule [NOTA-CTHRSSVVC] radiolabeled with the [68Ga]gallium and [18F]fluorine isotopes, for comparison. p-SCN-Bz-NOTA-CTHRSSVVC was prepared by the condensation reaction using the chelators p-Bn-SCN-DOTA, or p-SCN-Bz-NOTA, and the cyclic peptide CTHRSSVVC. The reaction yield, as well as the respective quality controls of the products, and their purifications were performed by high performance liquid chromatography (HPLC) and the peaks obtained were confirmed by mass spectrometry. In the case of radiolabeling, 80l of the conjugated peptide (NOTA-CTHRSSVVC), obtained commercially, was radiolabeled in 0.350 mL of 0.2 M sodium acetate solution (pH 4.1), with 0.05 mL containing 1.0 - 2.5 mCi (or 37 - 92.5 MBq) of [68Ga]Ga or [18F]AlF; the reactions were monitored under different temperatures and the quality controls of the radiolabeled products were evaluated by HPLC coupled to a gamma radiation detector. The preparation of p- SCN-NOTA-CTHRSSVVC, by condensation reaction, was analyzed by LC-MS and the structures were confirmed by the Q-TOF technique. The radiolabeling of conjugated peptides [68Ga] Ga-NOTA-CTHRSSVVC or [18F]AlF-NOTA-CTHRSSVVC, purified, were analyzed by HPLC producing products with radiochemical purity of 97.8% (n = 3) and 95.5% (n = 3), respectively. Both of the radioactive products were chemically stable for 4 hours
