O tabagismo é um importante fator de risco para várias doenças cardiovasculares e pulmonares, e também têm sido associado à perda de massa e força muscular esquelética levando à sarcopenia. Assim, o presente estudo analisou os efeitos histopatológicos do processo de envelhecimento do diafragma, em indivíduos de ambos os sexos, em diferentes faixas etárias, a fim de avaliar se a carga tabágica é responsável por acréscimo das correlações clinicopatológicas. Amostras do músculo diafragma post-mortem foram coletadas e processadas para análise histológica através das colorações histológicas de Picrosirius red e hematoxilina e eosina. Utilizou-se a estereologia e a morfometria para mensurar as alterações nas fibras musculares, bem como a densidade de volume das fibras colágenas, adipócitos e vasos sanguíneos (incluindo capilares). A análise estatística, com nível de significância P < 0,05, foi realizada através do programa SPSS. Os dados foram apresentados como média ± erro padrão da média para vaiáveis contínuas e frequência absoluta e relativa para as categóricas. Utilizamos o Teste T de Student (bicaudal e não pareado) para comparar as variáveis continuas entre os indivíduos fumantes com os não-fumantes (controle). As variáveis categóricas foram analisadas através do Teste Qui-quadrado ou Exato de Fisher quando necessário. Para testar se as distribuições de tamanhos de fibras normais eram diferentes entre os grupos, foi utilizado o teste de Kolmogorov-Smirnov. A fim de entender como as variáveis demográficas e clínicas podem atuar nos achados histopatológicos, realizamos uma análise de regressão multivariada. Observamos que com o avanço da idade, o diafragma acumula características histopatológicas que podem levar a complicações na mecânica respiratória. Ademais, o tabagismo demonstrou atuar negativamente na histologia diafragmática, sendo que a carga tabágica desempenha papel de suma importância no acréscimo de características histopatológicas neste músculo

Smoking is a major risk factor for several cardiovascular and pulmonary diseases, and has also been associated with loss of skeletal muscle mass and strength leading to sarcopenia. Thus, the present study analyzed the histopathological effects of the aging process of the diaphragm, in individuals of both sexes, in different age groups, in order to assess whether the smoking load is responsible for the increase in clinicopathological correlations. Post-mortem diaphragm muscle samples were collected and processed for histological analysis through Picrosirius red and hematoxylin and eosin staining techniques. Stereology and morphometry were used to measure changes in muscle fibers, as well as the volume density of collagen fibers, adipocytes and blood vessels (including capillaries). Statistical analysis, with a significance level of P < 0.05, was performed using the SPSS software. Data were presented as mean ± standard error of mean for continuous variables and absolute and relative frequency for categorical variables. We used Student's T test (two-tailed and unpaired) to compare continuous variables between smokers and non-smokers (control). Categorical variables were analyzed using the chi-square test or Fisher's exact test when necessary. To test whether normal fiber size distributions were different between groups, the Kolmogorov-Smirnov test was used. In order to understand how demographic and clinical variables may affect histopathological findings, we performed a multivariate regression analysis. We observed that with advancing age, the diaphragm accumulates histopathological characteristics that can lead to complications in respiratory mechanics. Furthermore, smoking has shown to have a negative effect on diaphragmatic histology, and smoking history plays a key role in the increase of histopathological characteristics in this muscle
