A maioria dos neurônios serotonérgicos no cérebro é contido em três grandes grupamentos diferentes, estão presentes nos núcleos dorsal da rafe (DR), no núcleo mediano (MnR) da rafe, e também na região supralemniscal, designada grupamento B9. O DR e MnR foram implicados em uma gama de funções fisiológicas, e particularmente em diferentes respostas fisiológicas relacionada ao estresse. Devido a uma organização difusa, quando comparado com o DR e MnR, pouco é conhecido sobre as conexões e funções do grupamento B9. Apenas poucos dados na literatura indicam que o DR, MnR e B9 são interconectados reciprocamente. Contudo, acumulam evidências de que esses núcleos, tradicionalmente considerados serotonérgicos, na verdade, são altamente heterogêneos, contendo populações de neurônios com fenótipos neuroquímicos, projeções, e propriedades eletrofisiológicas distintas. Porém, até o momento, o exato padrão das conexões intrínsecas entre o DR, MnR e B9, assim como o fenótipo neuroquímico dos neurônios dando origem a elas, ainda não foram investigados de forma sistemática. Desta forma, visamos nesse estudo, investigar através de métodos de rastreamento neural combinados com métodos de imunofluorescência, a exata topografia e o fenótipo neuroquímico das conexões entre DR, MnR e o grupamento B9. As injeções dos traçadores retrógrados e anterógrados no DR e MnR, revelaram que eles são interconectados de forma recíproca. Identificamos as asas laterais do DR (DRL) como a principal fonte das projeções do DR para o MnR e B9, como também, o principal alvo de axônios provenientes do MnR. Através da dupla-marcação para serotonina (5-HT) e para o transportador vesicular de glutamato do tipo 3 (VGLUT3), verificamos que no MnR e no grupamento B9, apenas 50% dos neurônios imunomarcados exibem um fenótipo serotonérgicos. As combinações de rastreamento retrógado com imunofluorescência para 5-HT e/ou VGLUT3 revelaram que 95% das aferências do DR para o MnR foi originada de neurônios não-serotonérgicos, não-glutamatérgicos nas asas laterais (DRL). Além disso, poucos neurônios glutamatérgicos, serotonérgicos, ou com um fenótipo misto, em outras sub-regiões do DR contribuíram para as projeções do DR para o MnR. Esses resultados indicam que DR, MnR e B9 são interconectadas de forma recíproca, e destacam o DRL como a principal fonte e alvo dessas projeções intrínsecas. E caracterizam ainda mais o DR, MnR e o B9 como grupamentos altamente heterogêneos, apresentando vários tipos de neurônios com fenótipos neuroquímicos distintos.

The majority of serotonergic neurons in the brain are confined to three large distinct cell groups, the dorsal (DR) and median (MnR) raphe nuclei, and a supralemmiscal region, termed B9 cell group. DR and MnR have been implicated in a vast array of physiological functions and specifically in distinct physiological responses to stress. Due to its more diffuse organization in comparison to DR and MnR, little is known about the connections and functions of the B9 cell group. There are only few data in the literature describing that DR, MnR, and B9 are interconnected reciprocally. Moreover, evidence is accumulating that these nuclei traditionally considered as serotonergic, in fact are highly heterogeneous, consisting of cell populations with distinct transmitter phenotypes, connections, and electrophysiological properties. However, till yet the exact pattern of the intrinsic connections between DR, MnR, and B9, as well as the transmitter phenotype of the neurons which give rise to them, were not investigated systematically. Thus, in the present study, we examined by neuronal tracing, combined with immunofluorescence techniques the exact topography and transmitter phenotype of the projections between DR, MnR, and B9. Injections of retrograde and anterograde tracers into the DR and MnR revealed that these regions are robustly interconnected reciprocally. We identified the lateral wings of the DR (DRL) as the principal source of DR projections to the MnR, and B9, as well as the primary target of axons arising from MnR. By double-staining for serotonin (5-HT) and the type 3 vesicular glutamate transporter (VGLUT3) we verified that only about 50% of the immunostained neurons exhibit a purely serotonergic phenotype. By combining retrograde tracing with immunofluorescence staining for 5-HT and/or VGLUT3 we revealed that about 95% of the DR afferents to MnR arose from non-serotonergic, non-glutamatergic DRL neurons, In addition, only few purely glutamatergic, purely serotonergic, or neurons with a mixed phenotype in other DR subregions contributed to DR projections to the MnR. These findings indicate that DR, MnR, and B9 are amply interconnected reciprocally and highlight the DRL as the principal source and target of these intrinsic connections. They also further characterize DR, MnR, and B9 as highly heterogeneous cell groups, which contain various types of neurons with distinct transmitter phenotypes.
