Um dos principais fatores causadores de distúrbios homeostáticos observados nas epilepsias, são as complicações observadas no sistema respiratório, em especial, as apneias, que provocam aumento dos níveis de CO2 (hipercapnia) e/ou diminuição dos níveis de O2 no sangue arterial (hipoxemia). Os neurônios respiratórios, localizados na coluna respiratória ventral (VRC), constituem um dos principais grupamentos responsáveis por controlar a atividade respiratória. Trabalhos recentes do nosso grupo de pesquisa demostraram alterações respiratórias em dois modelos experimentais de epilepsia, o modelo de epilepsia audiogênica (WAR) e o de estimulação rápida da amígdala (ARK). No presente estudo, procuramos avaliar alterações respiratórias no modelo clássico de epilepsia do lobo temporal (ELT) com uma abordagem mais seletiva, via injeção intra-hipocampal de pilocarpina. Foram utilizados ratos Wistar adultos (197-492 g; N = 4-9/grupo) com cânulas de aço inoxidável implantadas na região do hipocampo. Os parâmetros respiratórios analisados pela técnica de pletismografia de corpo inteiro foram a frequência respiratória (fR), volume corrente (VT) e ventilação-minuto (VE). Nesse mesmos animais, também avaliamos os parâmetros de mecânica respiratória como a resistência Newtoniana de vias aéreas, resistência ou viscância do parênquima pulmonar e a elastância do parênquima pulmonar. Quinze e trinta dias após a injeção de pilocarpina (2,4 mg/μl), observamos uma redução da resposta taquipneica frente a uma situação de hipercapnia (7% CO2). Além disso, não foram observadas alterações significantes na VE basal e durante estímulos hipóxicos (8% O2) trinta dias após o status epilepticus. No entanto, quinze dias após a injeção de pilocarpina, observamos um aumento significativo da ventilação frente ao desafio hipóxico, mas não em condições basais. Também não foram observadas alterações na resistência das vias aéreas, na viscância e na elastância do parênquima pulmonar. Dessa forma, no modelo de ELT, com a injeção de pilocarpina no hipocampo, apenas a resposta de aumento da fR foi atenuada, sugerindo que, pelo menos no período de até 30 dias, o padrão e ritmo respiratório apresentam-se intactos, mas o mecanismo de taquipneia parece estar comprometido.

One of the main causes of homeostatic disorders observed in epilepsy is the complications observed in the respiratory system, especially apneas, which cause an increase in CO2 levels (hypercapnia) and/or a decrease in O2 levels in arterial blood (hypoxemia). The respiratory neurons located in the ventral respiratory column (VRC) are one of the main groups responsible for controlling respiratory activity. Recent data from our group has demonstrated respiratory changes in two experimental models of epilepsy, audiogenic epilepsy (WAR) and rapid stimulation of the amygdala (ARK). Here, we aimed to evaluate respiratory changes in the classic model of temporal lobe epilepsy (TLE) with intra-hippocampal injection of pilocarpine. Adult Wistar rats (250-430 g; N = 4-7/group) with stainless steel cannulas implanted in the hippocampus region were used. The respiratory parameters analyzed by the whole-body plethysmography technique were respiratory rate (fR), tidal volume (VT) and ventilation (VE). In these same animals, we also evaluated the parameters of respiratory mechanics such as Newtonian airway resistance, resistance of the pulmonary parenchyma and the elasticity of the pulmonary parenchyma. Fifteen and thirty days after pilocarpine injection (2,4 mg/μl), we observed a reduction of the tachypneic response to hypercapnia (7% CO2). No significant changes were observed in basal VE and during hypoxic stimuli (8% O2). We also did not observe changes in airway resistance, pulmonary parenchymal resistance, and pulmonary parenchyma elasticity. Thus, in the TLE model with the injection of pilocarpine only the response of increased fR was attenuated, suggesting that, at least in the period of 30 days, the respiratory pattern and rhythm are intact, but the tachypneic mechanism seems to be affected.
