Individuos expostos a estresse estão em risco de desenvolver várias desordens mentais, incluindo o transtorno do estresse pós-tratumático (TEPT), uma condição debilitante que ocorre geralmente em comorbidade com outras condições psiquiátricas e, também, disfunções sexuais. Além de ser um transtorno psiquiátrico, o TEPT tem sido também associado a desregulação da imunidade inata, entre outros fatores metabólicos e inflamatórios. Embora o TEPT tem sido ligado a uma condição inflamatória sistêmica, pouco é conhecido dos efeitos celulares e molecular da imunidade inata no sistema nervoso central (SNC) e o impacto no ambiente póstesticular (epidídimo) e fertilidade masculina. No presente estudo, o objetivo foi avaliar as consequências do estresse por isolamento social (SI), que mimetiza no camundongo adulto a condição clínica de PTSD, em componentes celulares e moleculares da imunidade inata no SNC e epididimo. Os objetivos específicos foram: 1) caracterizar o padrão de expressão da b-defensina SPAG11C no SNA de animais controles; 2) avaliar is efeitos do IS no padrão de expressão e regulação de componentes celulares e moleculares da imunidade inata, incluindo a b-defensin SPAG11C, no SNC e em mediadores inflamatórios plasmáticos e teciduais; e 3) avaliar os efeitos do IS no perfil de expressão e regulação de componentes celulares e moleculares da imunidade inata, incluindo a b-defensina SPAG11C, no epidídimo e em parâmetros espermáticos. Em conjunto, os resultados apontam para novos aspectos do entendimento do papel de componentes da imunidade inata na neuroinflamação do TEPT e como esta condição clinica impacta a imunobiologia epididimária levando a uma epididimite que, então, afeta negativamente parâmetros espermáticos. Ainda, o estudo dá pistas de possíveis novos papéis biológicos para a b-defensina SPAG11C em células imunológicas e não imunológicas no SNC e epidídimo em condição de homeostasia e inflamatória.

Individuals are exposed to stress develop a range of mental disorders, including posttraumatic stress disorder (PTSD), a debilitating condition usually in comorbidity with other psychiatric disorders and sexual dysfunction. In addition to psychiatric impairment, PTSD has been associated with dysregulation of innate immunity, among other metabolic and inflammatory factors. Although PTSD has been linked to a systemic inflammation condition, little is known about how its effects on cellular and molecular aspects of innate immunity in the central nervous system (CNS) and impact on the post-testicular environment (epididymis) and male fertility. In the present study, we aimed to evaluate the consequences of stress by social isolation (SI), which mimics in adult mice with the PTSD clinical condition, on cellular and molecular components of innate immunity in the CNS and epididymis. The specific aims were: 1) characterize the expression pattern of the b-defensin SPAG11C in the CNS of control mice; 2) evaluate the effects of SI the expression profile and regulation of innate immunity cellular and molecular components, including b-defensin SPAG11C on the CNS as well as plasma and tissue inflammatory mediators; and 3) evaluate the effects of SI the expression profile and regulation of innate immunity cellular and molecular components, including b-defensin SPAG11C on the epididymis and sperm parameters. Altogether, the results point out to new insights into the understanding of the role of innate immunity components in PTSD neuroinflammation and how this clinical condition impacts epididymal immunobiology leading to an epididymitis condition that negatively affects sperm parameters. Furthermore, the study potentially unravels new roles for the b-defensin SPAG11C in immune and non-immune cells in the CNS and epididymis in homeostasis and inflammatory conditions.
