Nas últimas décadas, diversos estudos demonstraram a importante ação anti-inflamatória da CTX, avaliada diretamente sobre funções de neutrófilos e macrófagos, além do seu potencial como imunomodulador, particularmente sobre macrófagos relacionados à progressão tumoral e resposta inflamatória aguda e infecciosa. Esta capacidade imunomoduladora, de longa duração (até 14 dias) da CTX envolve a reprogramação fenotípica de macrófago, tornando-o mais pró- ou anti-inflamatório, dependendo da concentração da toxina e do estímulo do microambiente. Particularmente, com a presença local de macrófagos a produção e liberação dos mediadores químicos produzidos por eles, a migração e ativação de fibroblastos é intensificada, levando à deposição de grandes quantidades de fibronectina e colágeno, o que pode favorecer a fibrose. Portanto, o objetivo do presente projeto foi investigar a importância da atividade secretora de macrófagos induzida pela CTX sobre as funções de fibroblasto envolvidas com o processo de cicatrização. Para tanto, monoculturas de macrófagos diferenciados a partir da linhagem monocítica humana THP-1 foram incubados com diferentes concentrações da CTX, por 2 horas, lavados e incubados com meio RPMI 1640 fresco, por 24 horas e após esse período, fibroblastos humanos foram incubados com os sobrenadantes dessas monoculturas (cel:sobrenadante) ou com os macrófagos (cel:cel) para a determinação in vitro dos eventos dos fibroblastos, em diferentes períodos, tais como: 1) ensaio de proliferação; 2) ensaio de migração no modelo de Wound healing; 3) ensaio de migração em Câmara de transwell; 4) morfologia do citoesqueleto de actina dos fibroblastos e expressão de fibronectina e colágeno tipo I secretados por essas células, visualizados por microscopia confocal; 5) quantificação de 15-Epi-LXA4, por ensaio imunoenzimático (EIA) e 6) Análises proteômica e secretômica de macrófagos tratados com a toxina, por espectrometria de massas. Em conjunto, os resultados apresentados mostram que macrófagos tratados com CTX modulam a atividade funcional de fibroblastos (migração e proliferação e secreção de componentes de matriz), tanto no contato célula:célula, como por meio de mediadores secretados em seus sobrenadantes, de maneira mais pronunciada ao observado para anti-inflamatórios clássicos. Esse estudo contribuiu, portanto, para evidenciar a importância da capacidade imunomoduladora da CTX sobre eventos envolvidos com a cicatrização.

In the last decades, several studies have demonstrated the important anti-inflammatory action of CTX, directly assessed on the functions of neutrophils and macrophages, in addition to its potential as an immunomodulator, particularly on macrophages related to tumor progression and acute and infectious inflammatory response. This long-term immunomodulatory capacity (up to 14 days) of CTX involves phenotypic reprogramming of the macrophage, making it more pro- or anti-inflammatory, depending on the concentration of the toxin and the microenvironment stimulus. Particularly, with the local presence of macrophages, the production and release of the chemical mediators produced by them, the migration and activation of fibroblasts is intensified, leading to the deposition of large amounts of fibronectin and collagen, which can favor fibrosis. Therefore, the objective of the present project was to investigate the importance of macrophage secreting activity induced by CTX on the fibroblast functions involved in the healing process. Therefore, monocultures of macrophages differentiated from the human monocytic strain THP-1 were incubated with different concentrations of CTX, for 2 hours, washed and incubated with fresh RPMI 1640 medium, for 24 hours and after that period, human fibroblasts were incubated with the supernatants of these monocultures (cel: supernatant) or with macrophages (cel: cel) for the in vitro determination of fibroblast events, in different periods, such as: 1) proliferation assay; 2) migration test in the Wound healing model; 3) migration test in the Transwell Chamber; 4) morphology of the fibroblast actin cytoskeleton and expression of fibronectin and type I collagen secreted by these cells, visualized by confocal microscopy; 5) quantification of 15-Epi-LXA4, by immunoenzymatic assay (EIA) and 6) Proteomic and secretomic analyzes of macrophages treated with the toxin, by mass spectrometry. Together, the results presented show that macrophages treated with CTX modulate the functional activity of fibroblasts (migration and proliferation and secretion of matrix components), both in cell:cell contact, and through mediators secreted in their supernatants, in a more efficient way. pronounced to that observed for classic anti-inflammatory drugs. This study, therefore, contributed to highlight the importance of immunomodulatory capacity of CTX on events involved with healing.
