A doença de Parkinson (DP) é uma desordem neurodegenerativa associada à perda de neurônios dopaminérgicos (DA) na porção compacta da substância negra (SNc). No entanto, outros neurônios do tronco encefálico também podem estar degenerados. O objetivo do presente trabalho foi correlacionar as alterações neuroanatômicas e biomoleculares às alterações respiratórias observadas no modelo experimental para DP. Ainda, observar se a ativação farmacológica ou optogenética é capaz de restaurar a função respiratória destes animais. Foram utilizados ratos Wistar adultos e o modelo experimental foi de injeção bilateral da toxina 6-OHDA nas regiões dos núcleos Caudado e Putâmen (CPu). Os parâmetros respiratórios foram avaliados por pletismografia, além da análise eletrofisiológica dos músculos respiratórios. Foram realizadas injeções bilaterais de NMDA ou unilaterais do promotor viral PRSx8 no RTN a fim de verificar se a ativação destes neurônios poderia restaurar a função respiratória neste modelo. Os resultados mostraram que os animais que receberam 6-OHDA, apresentaram redução no número de neurônios em importantes regiões bulbares associados ao controle da respiração, como o núcleo retrotrapezóide (RTN). As análises biomoleculares demonstraram haver um remodelamento proteico na região do RTN após a indução do modelo. Foi possível observar redução na frequência respiratória (fR) e ventilação (VE) 40 e 60 dias após a lesão. A estimulação farmacológica no RTN em ratos não anestesiados, promoveu aumento na ventilação. A estimulação optogenética em animais anestesiados ou não, resultou em respostas maiores nos animais do grupo experimental. Concluímos que a injeção de 6-OHDA no CPu promove alterações neuroanatômicas e biomoleculares que precedem as funcionais e que a estimulação farmacológica e optogenética é capaz de aumentar a ventilação nos animais submetidos ao modelo experimental para DP. Os resultados obtidos com a combinação de técnicas de proteômica e optogenética, poderão abrir caminho para futuras linhas de pesquisa associadas às doenças neurodegenerativas.

Parkinson's disease (PD) is a neurodegenerative disorder associated with loss of dopaminergic neurons (DA) in the compact portion of the substantia nigra (SNc). However, other brainstem neurons may also be degenerate. The aim of the present study was to correlate the neuroanatomic and biomolecular changes to the respiratory changes observed in the experimental model for PD. Also, observe whether the pharmacological or optogenetic activation is able to restore the respiratory function of these animals. Adult Wistar rats were used and the experimental model was bilateral injection of the 6-OHDA toxin in the Caudado and Putâmen (CPu) regions. The respiratory parameters were evaluated by plethysmography, in addition to the electrophysiological analysis of the respiratory muscles. Bilateral NMDA or unilateral injections of the PRSx8 viral promoter into the RTN were performed to verify whether the activation of these neurons could restore respiratory function. The results showed that animals receiving 6-OHDA showed a reduction in the number of neurons in important regions associated with respiratory control, such as the retrotrapezoid nucleus (RTN). The biomolecular analyzes showed a protein remodeling in the region of the RTN after the induction of the model. Reduction in respiratory rate (fR) and ventilation (VE) 40 and 60 days after injury were observed. Pharmacological stimulation in the RTN in non- anesthetized rats promoted an increase in ventilation. Optogenetic stimulation in anesthetized or non-anesthetized animals resulted in larger responses in experimental group animals. We conclude that the 6-OHDA injection in CPu promotes neuroanatomical and biomolecular alterations that precede the functional alterations and that the pharmacological and optogenetic stimulation is able to increase the ventilation in the animals submitted to the experimental model for PD. The results obtained in this work with the combination of proteomics and optogenetic techniques, can open the way for future lines of research associated with neurodegenerative diseases.
