Ouriços do mar são equinodermos bentônicos amplamente distribuídos geograficamente, que possuem um sistema imune inato bem desenvolvido capaz de fornecer uma ampla gama de respostas celulares e moleculares. As funções de seu sistema imune estão diretamente relacionadas aos celomócitos, diferentes tipos celulares presentes no líquido celomático. Dentre estas células, os esferulócitos vermelhos são considerados mediadores da resposta imune por meio da atividade antioxidante e bactericida do pigmento equinocromo-A presente em seus grânulos citoplasmáticos. Apesar da atividade descrita para os esferulócitos, ainda pouco se sabe sobre os mecanismos de atuação deles no sistema imune inato de ouriços do mar. Diferentes estudos mostram o aumento da proporção dos esferulócitos vermelhos em diferentes condições de estresse ambiental. Diante disso, este trabalho teve como objetivo identificar o pigmento presente nos esferulócitos vermelhos do ouriço do mar antártico Sterechinus neumayeri, bem como avaliar suas funções no sistema imune inato. O pigmento foi extraído a partir dos celomócitos, purificado por cromatografia líquida de alta eficiência em fase reversa (RP-HPLC), submetido a análises por espectrometria de massas (ESI-MS) e posteriormente avaliada sua atividade biológica em um contexto imunológico e molecular. Foram realizados ensaios in vitro com duas concentrações do pigmento (50 e 100µg/mL) durante 1h, 6h e 24 horas de incubação. Avaliou-se também o efeito dele na expressão dos genes policetídeo sintase (Pks), sulfotransferase (SULT), superóxido dismutase (SOD), catalase (CAT), metalotioneína (MT) e Hsp70. O tratamento das células com diferentes concentrações de pigmento não alterou a expressão de PKs, SULT e MT. Já para SOD, CAT e Hsp70 observou-se a diminuição da expressão em 1 hora a 100 µg/mL, seguida de aumento da expressão de CAT em 24 horas, para ambas as concentrações testadas. Constatou-se que os esferulócitos vermelhos de S. neumayeri possuem um novo pigmento naftoquinônico polihidroxilado. Além disso, verificou-se que esta espécie não apresenta o equinocromo-A. Observou-se aumento da capacidade fagocítica, em relação ao controle, após uma 1 hora de tratamento com as duas concentrações do pigmento. A análise da morfologia dos amebócitos fagocíticos após 1 hora de incubação com o pigmento mostrou aumento na marcação do citoesqueleto de actina, provavelmente devido a intensificação da polimerização dos filamentos de actina. Além disso foi observado uma diminuição da área dos amebócitos fagocíticos. Conclui-se que o pigmento presente nos esferulócitos vermelhos de S. neumayeri seja um forte candidato ao papel de imunoestimulador de amebócitos fagocíticos para esta espécie de ouriço do mar.

Sea urchins are widely geographically distributed benthic echinoderms, which have a well-developed innate immune system capable of providing a wide range of cellular and molecular responses. All functions of your immune system are directly related to the coelomocytes, different cell types present in coelomic fluid. Among these cells, red sphere cells are considered mediators of immune response through a pigment with antioxidant and bactericidal activity, the echinochrome-A, located inside cytoplasmic granules. Despite the activities described for sphere cells, not so much is known about mechanisms of their action in the innate immune system of sea urchins. Different studies show an increased proportion of red sphere cells under different conditions of environmental stress. This study aimed to identify the pigment present in the sphere cells of antarctic sea urchin Sterechinus neumayeri as well as evaluate their functions in the innate immune system. The pigment was extracted from coelomocytes, purified by reverse phase high performance liquid chromatography (RP-HPLC), subjected to mass spectrometric analysis (ESI-MS) and then its biological activity was evaluated in the context of cell immune response. In vitro assays were performed with two pigment concentrations (50 and 100µg / mL) for 1h, 6h and 24 hours of incubation time. The pigment`s effect on gene expression was also evaluated for polyketide synthase (Pks), sulfotransferase (SULT), superoxide dismutase (SOD), catalase (CAT), metallothionein (MT) and Hsp70. Treatment of cells with different concentrations of pigment did not alter the expression of PKs, SULT and MT. For SOD, CAT and Hsp70 decreased expression at 1 hour at 100 µg /mL was observed, followed by 24-hour CAT expression increase for both tested concentrations. A new polyhydroxylated naphthoquinone pigment was found in red sphere cells of S. neumayeri. In addition, the echinochrome-A was not found in antarctic species. It was observed increased in phagocytic capacity compared to control after 1 hour treatment with both pigment concentrations. The analysis of morphology of phagocytic amoebocytes after 1 hour incubation with the pigment showed an increase in actin cytoskeleton labeling, probably due to the intensification of the actin filaments polymerization. In addition, a decrease in phagocytics amebocytes area were observed. We can conclude that the pigment present in S. neumayeri red sphere cells is a strong candidate for the role of phagocytic amebocyte immunostimulator in this species of sea urchin.
