Moléculas de origem animal com atividade imunomoduladora tem sido alvo de inúmeros estudos devido seu potencial terapêutico. O envenenamento pela cascavel Crotalus durissus terrificus apresenta como características a baixa resposta inflamatória local, porém ações sistêmicas. Somado a isso, este veneno induz baixa produção de anticorpos contra seus componentes, assim como contra um antígeno heterólogo. A crotoxina (CTX) é o componente majoritário deste veneno e, dentre suas atividades biológicas está a ação imunomoduladora. Nós temos estudado o efeito modulador da CTX sobre a imunidade adaptativa. Com relação à imunidade humoral, em estudo anterior foi verificado que a administração da CTX antes ou após a imunização de camundongos com a albumina humana (SAH) na presença de adjuvantes distintos resulta na inibição da produção de anticorpos anti-SAH. Considerando que os linfócitos B são responsáveis pela produção dos anticorpos antígeno-específicos e pela memória imunológica humoral, neste projeto estudamos o efeito da CTX sobre a produção de anticorpos anti-ovalbumina (OVA) e sobre os linfócitos B in vivo, assim como a ativação dessa população celular in vitro. Para isso, avaliamos a produção de anticorpos anti-OVA em diferentes períodos após a imunização dos camundongos com OVA adsorvida em Alum e que receberam ou não a CTX e ainda, as populações de linfócitos B presentes nos linfonodos drenantes do sítio de imunização desses animais. Além disso, estudamos o efeito direto da CTX sobre a ativação e proliferação das células B incubadas com estímulos mitogênicos in vitro. Os resultados permitiram verificar que a administração da CTX resultou em menor produção de anticorpos IgM, IgG1 e IgG2a anti-OVA em relação ao obtido nos animais apenas imunizados com a OVA, além disso verificamos menor porcentagem das populações de células B. Os resultados obtidos sugerem também que a CTX inibe a população de células CD4+CXCR5+ nos animais imunizados com OVA. Por outro lado, verificamos que a produção de anticorpos anti-OVA em animais imunizados 90 dias antes e que receberam ou não a CTX foi semelhante. Em experimentos in vitro, verificamos que a CTX não induz morte dos linfócitos B, no entanto, foi capaz de inibir a proliferação de células B incubadas com LPS na presença ou não de anti-CD40 ou ainda, incubadas com anticorpos anti-IgM e anti-CD40. Observamos também que a CTX inibiu a expressão das moléculas CD80 e CD86 nos linfócitos B incubados com LPS e anti-CD40. Além disso, verificamos que a CTX potencializou a expressão de IL-10 em células CD19+ incubadas com LPS in vitro. Portanto, o conjunto de resultados obtidos demonstra que a CTX é capaz de atuar diretamente sobre os linfócitos B interferindo na sua ativação, proliferação e produção de anticorpos frente a um estímulo heterólogo como a OVA ou mitogênico.

Molecules of animal origin that present immunomodulatory activity have been the focus of several scientific studies due to their therapeutic potential. The envenoming by the Crotalusdurissus terrificus (C.d.terrificus) rattlesnake is characterized by low local inflammatory reaction in addition to the systemic effects. In addition, it was shown that, distinct from others snake venoms, the C.d.terrificus venom induces low antibodies production against its components, as well as against an unrelated antigen. The crotoxin (CTX) is the main component of this venom and, among its biological activities, it is included the immunomodulatory action. We have been studied the modulatory effect of CTX on adaptive immunity. Concerning about the humoral immunity, we previously showed that the CTX administration before or after the mice immunization with human serum albumin (HSA) in the presence of distinct adjuvants results in inhibition of anti-HSA antibody production. Considering that the B lymphocytes are responsible by antigen-specific antibody production and also humoral immunological memory, in this project we studied the effect of CTX on the antibody production induced by ovalbumin (OVA) as well as on B lymphocytes activation in vivo and in vitro. For this, we evaluated the production of anti-OVA antibodies in different periods after the immunization of mice with OVA adsorbed in Alum and that received or not the CTX as well as the B cell populations present in the draining lymph nodes of the immunization site of these mice. Furthermore, we studied the direct effect of CTX on B cells activation and proliferation induced by mitogenic stimulus in vitro. The results showed that the administration of CTX resulted in lower production of IgM, IgG1 and IgG2a anti-OVA antibodies compared to those obtained in mice only immunized with OVA accompanied by lower percentage of all B cell populations evaluated. The data also suggest that CTX inhibits the CD4+CXCR5+ cell population in mice immunized with OVA. On the other hand, similar amounts of anti-OVA antibody production in mice immunized 90 days before and that received or not CTX. In in vitro experiments, we observed that CTX does not induce death of B lymphocytes, as well as the proliferation of these cells. In addition, we found that CTX was able to inhibit the proliferation of B cells incubated with LPS in the presence of anti-CD40 mAb or incubated with anti-IgM and anti-CD40 mAbs. We also observed that the CTX inhibited the expression of CD80 and CD86 molecules in B lymphocytes incubated with LPS and anti-CD40. In addition, we found that CTX potentiated IL-10 expression in CD19+ cells incubated with LPS in vitro. Therefore, our results demonstrate that the CTX is able to act directly on B lymphocytes interfering in their activation, proliferation and production of antibodies induced by a heterologous antigen and also mitogenic stimulation.
