O glioblastoma (GBM) é o tumor maligno mais frequente do Sistema Nervoso Central, possui um prognóstico ruim, perspectiva de vida abaixo de 15 meses e aumento em sua incidência todos os anos. O tratamento atual consiste em ressecção cirúrgica seguida de radioterapia e quimioterapia com Temozolomida, mesmo assim essa estratégia demonstra-se falha pelo alto índice de reincidência tumoral e baixa sobrevida dos pacientes. Assim, outras abordagens terapêuticas vêm sendo desenvolvidas e, dentre elas, a imunoterapia. Considerando-se que no GBM, os pacientes recebem, por padrão, tratamento com a Temozolomida, as pesquisas concentram seus esforços na associação da quimioterapia com outras formas de tratamento, especialmente a imunoterapia. O presente projeto teve como objetivo investigar os efeitos da TMZ sobre o fenótipo das DCs derivadas de monócitos (Mo-DCs), que são usadas em diferentes protocolos de imunoterapia com vacinação de DCs. Para tanto, a TMZ foi adicionada sobre monócitos (simultaneamente à sua exposição as citocinas que induzem sua diferenciação em DC) ou sobre Mo-DCs imaturas (cinco dias após o início cio da cultura) e a cultura celular continuou até o sétimo dia para avaliação do seu fenótipo. A expressão de marcadores associados ao poder imunoestimulador das DC, CD80, CD86 e CD83 apresentou mudanças sugestivas de maturação de DCs imaturas, quando tratadas com o fármaco. Este efeito maturador da TMZ aparenta ser, inclusive maior que a citocina usualmente usada para induzir a maturação das DC, o TNF- α. Embora preliminares, também pareceu ocorrer aumento na produção de espécies reativas de oxigênio (EROS) nas células tratadas com TMZ isolada, que acabam causando lesões no DNA e a reparação está relacionada com a 10 mudança no perfil fenotípico e funcional das DCs, o que seria coerente com dados da literatura e com a atividade aparentemente maturadora da TMZ sobre as DCs imaturas vistas neste trabalho.

The Glioblastoma is the most common malignant tumor of the Cetral Nervous System. Has a bad progonosis, a life expectancy of less than 15 months and increase in it is incidence every year.
