A psoriase e uma doenca de carater inflamatorio que ocorre na pele, sendo caracterizada pelo desenvolvimento de eritemas derivados do processo inflamatorio cronico e a hiper proliferacao de queratinocitos na epiderme. Esta doenca afeta cerca 2-3% da populacao mundial e a qualidade de vida dos individuos que a portam, sendo mantidos em medicacao constante durante o aparecimento dos sintomas. Alem dos linfocitos Th17, queratinocitos participam da progressao da doenca atraves da sua hiperproliferacao e secrecao de citocinas. O papel dos queratinocitos na psoriase ainda nao e total compreendido e, nesse contexto, alguns estudos tem sugerido o metabolismo mitocondrial desta celula como um modulador da resposta imune durante a progressao da psoriase. A mitocondria e uma organela dinamica que esta em processo constante de fissao e fusao, o qual foi recentemente atrelado ao desenvolvimento de respostas inflamatorias. Neste contexto nosso grupo explorou o metabolismo mitocondrial durante a progressao da doenca e hipotetizou que a dinamica mitocondrial esta de queratinocitos impacta negativamente a progressao da doenca. Neste contexto, objetivamos estudar em modelos humanos in silico e de forma experimental em modelos in vitro e in vivo esta hipotese. Para a conducao de analises em humanos nos reanalisamos datasets de transcriptomica publicos. Em seguida, buscamos verificar os mecanismos em modelos experimentais in vitro e in vivo, nos quais utilizamos tratamentos com Mdivi-1 para verificar o perfil inflamatorio e mitocondrial, visando estabelecer uma conexao entre estes dois parametros. Nossos resultados demonstram que, durante a progressao da doenca, as mitocondrias apresentam um perfil disfuncional e tendem a se fundirem em queratinocitos em modelos murinos. Alem disso, estas celulas parecem aumentar sua biogenese mitocondrial nos grupos psoriaticos tanto em humanos quanto em modelos murinos de forma que este processo esta relacionado com o perfil inflamatorio destas celulas. Nossos dados contribuem para a caracterizacao metabolica do perfil mitocondrial de queratinocitos e demonstra sua associacao com a inflamacao presente na psoriase.

Role of Mitochondrial Dynamics in Keratinocytes and Its Impact on the Development of the Inflammatory Response in Experimental Psoriasis Models

Psoriasis is an inflammatory disease that occurs on the skin, and is characterized by the development of erythema derived from the chronic inflammatory process and the hyperproliferation of keratinocytes in the epidermis. This disease affects around 2-3% of the world's population and the quality of life of individuals who suffer from it, being kept on constant medication during the onset of symptoms. In addition to Th17 lymphocytes, keratinocytes participate in disease progression through their hyperproliferation and secretion of cytokines. The role of keratinocytes in psoriasis is not yet fully understood and, in this context, some studies have suggested the mitochondrial metabolism of this cell as a modulator of the immune response during the progression of psoriasis. Mitochondria are a dynamic organelle that is in a constant process of fission and fusion, which has recently been linked to the development of inflammatory responses. In this context, our group explored mitochondrial metabolism during disease progression and hypothesized that mitochondrial dynamics of keratinocytes negatively impacts disease progression. In this context, we aim to study this hypothesis in human in silico models and experimentally in in vitro and in vivo models. To conduct analyzes in humans we reanalyzed public transcriptomics datasets. Next, we sought to verify the mechanisms in in vitro and in vivo experimental models, in which we used treatments with Mdivi-1 to verify the inflammatory and mitochondrial profile, aiming to establish a connection between these two parameters. Our results demonstrate that, during disease progression, mitochondria have a dysfunctional profile and tend to fuse into keratinocytes in murine models. Furthermore, these cells appear to increase their mitochondrial biogenesis in psoriatic groups in both humans and murine models, so that this process is related to the inflammatory profile of these cells. Our data contribute to the metabolic characterization of the mitochondrial profile of keratinocytes and demonstrate its association with the inflammation present in psoriasis.
