A paracoccidioidomicose, uma micose sistêmica e endêmica em diversos países da América Latina, é um importante problema de saúde pública. Embora exista tratamento, o mesmo é prolongado e casos de recidiva são comuns. Nesse sentido, o desenvolvimento de novas abordagens terapêuticas, como as vacinas, são importante. Além disso, é imprescindível compreender a participação de algumas células na resposta vacinal. Desse modo, o objetivo dessa tese foi comprovar se os neutrófilos são importantes para a resposta vacinal utilizando modelo experimental de paracoccidioidomicose. Nós observamos que a vacina terapêutica reduziu a carga fúngica pulmonar em animais com 15 ou 30 dias de infecção. Além disso, foi observado redução de neutrófilos e formação de NETs nos pulmões dos animais infectados por 15 dias. Assim, nós concluímos que a vacina pode alterar a infiltração dos neutrófilos no local da infecção e isso parece depender do tempo de infecção. Além disso, nós observamos que os neutrófilos parecem ter um envolvimento durante o momento em que a vacina é administrada.

Abstract in English
Paracoccidioidomycosis, a systemic mycosis and endemic in several Latin American countries, is an important public health problem. Although there is treatment, it is prolonged and cases of relapse are common. In this sense, the development of new therapeutic approaches, such as vaccines, are important. In addition, it is imperative to understand the participation of some cells in the vaccine response. Thus, the aim of this thesis was to verify if neutrophils are important for the vaccine response using an experimental model of paracoccidioidomycosis. We observed that the therapeutic vaccine reduced lung fungal load in animals with 15 or 30 days of infection. In addition, neutrophil reduction and NET formation were observed in the lungs of infected animals for 15 days. Thus, we conclude that the vaccine may alter the infiltration of neutrophils at the site of infection and this seems to depend on the time of infection. In addition, we note that neutrophils appear to be involved during the time the vaccine is administered.
