A dengue é a arbovirose de maior impacto na saúde pública mundial. A doença é causada pelo vírus Dengue (DENV), o qual compreende quatro sorotipos antigenicamente distintos do vírus, como consequência a infecção por um deles não implica em proteção contra os sorotipos heterólogos. O diagnóstico clínico diferencial da dengue é difícil, por isso o diagnóstico laboratorial aparece como uma importante ferramenta para a confirmação dos casos suspeitos. Os testes sorológicos na plataforma de ELISA são amplamente utilizados na confirmação laboratorial da dengue, pois são sensíveis e de custo acessível. No entanto, apresentam especificidade comprometida, particularmente, em regiões onde circulam outras arboviroses. Nesse sentido, o objetivo desta proposta foi desenvolver estratégias de diagnóstico sorológico específico da dengue na plataforma de ELISA utilizando proteínas recombinantes baseadas em um fragmento da NS1 (nomeadas ΔNS1 dos DENVs 1-4). Os antígenos foram obtidos após expressão em sistema procarioto utilizando linhagens de Escherichia coli e purificados por cromatografia de afinidade. As proteínas recombinantes purificadas foram reconhecidas por soros de animais infectados pelos DENVs, sugerindo que as mesmas conservam epítopos conformacionais. As ΔNS1 dos DENVs 1-4 foram utilizadas em combinação como antígenos de fase sólida e permitiram a detecção de anticorpos gerados em animais infectados com os DENVs, sem a interferência de anticorpos gerados após infecção pelo ZIKV. Além disso, as ΔNS1 dos DENVs 1, 2 e 4 apresentaram maior reatividade com o soro do DENV homólogo. Os resultados obtidos são relevantes ao apresentarem novos imunobiológicos com antigenicidade, imunogenicidade preservadas e contribuem para o desenvolvimento de novas estratégias de diagnóstico sorológico específico para dengue.

Dengue is the arbovirose with larger impact on public health worldwide. The disease is caused by the Dengue virus (DENV), that comprises four antigenically distinct serotypes of the virus in such a way that infection by one then does not imply protection against the heterologous serotypes. The clinical diagnosis of dengue is hard and therefore the laboratory diagnosis is an important tool to confirm of suspected cases. Serological tests on the ELISA platform are widely used for dengue infection diagnosis, because they are sensitive and affordable. However, their specificity is compromised particularly in regions where others arboviruses prevail. Therefore, the aim of this study was to develop specific serological diagnostic strategies for dengue in the ELISA platform using recombinant proteins developed from a fragment of NS1 protein (named ΔNS1 of the DENVs 1-4). The antigens were obtained after expression in prokaryotic system using Escherichia coli strains and purified by affinity chromatography. Importantly, the purified recombinant proteins were recognized by sera from animals infected by DENVs, suggesting that ΔNS1 antigens have their conformational epitopes preserved. Moreover, the ΔNS1 of DENVs 1-4 were used in combination as solid phase antigens and allowed the detection of DENV in the sera from infected animals, without cross reaction with antibodies after ZIKV infection. In addition, the ΔNS1 of the DENVs 1, 2 and 4 showed higher reactivity with the sera of the homologous DENV, thus indicating their potential as tools to specifically identify DENV sorotypes. The results are relevant and present novel immunobiologicals with preserved antigenicity, immunogenicity and can contribute to the development of new serological diagnostic strategies specific for dengue infection.
