A família Togaviridae compreende um grande e diverso grupo de vírus responsável por surtos recorrentes em humanos. Dentre os vírus desta família, o vírus Chikungunya (CHIKV) é um alphavirus, importante em termos de morbidade, mortalidade e impacto econômico em diferentes regiões do mundo. Este estudo teve como objetivo realizar um mapeamento detalhado de epitopos das proteínas estruturais de CHIKV reconhecidos pelos anticorpos da classe IgG do soro de pacientes em fase de convalescência. As amostras de soro utilizadas foram coletadas no estado de Sergipe,2016. Com os resultados obtidos no mapeamento foram analisados peptídeos lineares sintéticos de células B correspondentes aos epitopos das proteínas estruturais E2 e E3 de CHIKV pela técnica de Elisa indireto à base de peptídeos. A proteína E2 foi o principal alvo da resposta imunológica onde foram identificados 3 epitopos conservados correspondentes aos Peptídeos P3, P4 e P5. Esses epitopos foram os mais reativos e específicos entre 11 epitopos analisados, e apresentaram potencial para utilização em ensaios de diagnóstico sorológico e desenvolvimento e ou melhoramento de vacinas para o vírus Chikungunya.

The Togaviridae family comprises a large and diverse group of viruses responsible for recurrent outbreaks in humans. Among the viruses of this family, the Chikungunya virus (CHIKV) is an alphavirus, important in terms of morbidity, mortality and economic impact in different regions of the world. This study aimed to perform a detailed epitope mapping of the CHIKV structural proteins recognized by the IgG class antibodies from patients in the convalescence phase. The serum samples used were collected in the state of Sergipe, Brazil, in 2016. Synthetic linear peptides corresponding of CHIKV E2 and E3 proteins epitopes were analyzed using a peptide based indirect Elisa in order to identify B cell epitopes. E2 protein was found as the main target of the immune response, and 3 conserved epitopes have been identified. The peptides P3, P4 and P5 were the most reactive and specific epitopes among 11 epitopes analyzed and presented potential for use in serological diagnostic tests and development and / or improvement of vaccines for the Chikungunya virus.
