Zika virus, ZIKV, foi descoberto em 1947 em Uganda durante uma pesquisa de campo sobre Febre Amarela. Porém, somente em 2016 tornou-se emergência global com o aparecimento de fetos e recém-nascidos com predominância de más formações congênitas neuronais de mães infectadas por ZIKV. Por ser única arbovirose descrida com transmissão vertical, entre mãe e feto, os mecanismos de transmissão durante a gestação são pouco conhecidos. O vírus pode ser encontrado no líquido amniótico e no feto. A placenta é uma barreira natural e eficaz contra microrganismos em geral. Porém, existe um grupo de organismos patogênicos capazes de atravessar essa barreira, infectar o feto e causar-lhes sequelas permanentes e até a sua morte ainda no útero ou durante ou após o nascimento. O ZIKV foi incluído pela Organização Mundial da Saúde no grupo de doenças infecciosas, TORCH, transmitidas durante a gravidez. A presente pesquisa visa descrever em células placentárias de primeiro trimestre de gestação, ex vivo, a replicacão do arbovírus, e comparar o comportamento das linhagens Africana e Asiático-Brasileiro em tais células. Da linhagem Africana, até o momento desconhece sua transmissão vertical tenha causado as patologias apresentadas pela linhagem Asiático-Brasileiro. Porém, ambas são capazes de infectar células placentárias BeWo e HTR-8 em ex vivo. A hipótese visa testar se a linhagem Asiático-Brasileiro é mais virulento do que a Africana. Os resultados da dinâmica da cinética do ZIKV Africano apresentam ser mais virulento do que Asiático-Brasileiro. Esse, porém, permanece por mais tempo nas células de modo a sustentar a infecção por um período mais prolongado. Tal condição pode prolongar a infecção durante a gestação e consequentemente atravessa a barreira placentária e infectar o feto.

Zika virus, ZIKV, was discovered in 1947 in Uganda during a field research on Yellow Fever. However, it was only in 2016 that it became a global emergency with the appearance of fetuses and newborns with a predominance of neuronal congenital malformations of mothers infected with ZIKV. As it is the only arbovirus infection described with vertical transmission, between mother and fetus, the transmission mechanisms during pregnancy are poorly understood. The virus can be found in the amniotic fluid and in the fetus. The placenta is a natural and effective barrier against microorganisms in general. However, there is a group of pathogenic organisms capable of crossing this barrier, infecting the fetus and causing permanent sequelae and even death in the womb or during or after birth. ZIKV was included by the World Health Organization in the group of infectious diseases, TORCH, transmitted during pregnancy. The present research aims to describe in placental cells of the first trimester of gestation, ex vivo, the replication of arbovirus, and to compare the behavior of African and Asian-Brazilian strains in such cells. Of the African lineage, so far it is unknown that its vertical transmission has caused the pathologies presented by the Asian-Brazilian lineage. However, both are able to infect BeWo and HTR-8 placental cells in ex vivo. The hypothesis aims to test whether the Asian-Brazilian lineage is more virulent than the African. The results of the dynamics of the African ZIKV's kinetics appear to be more virulent than Asian-Brazilian. This, however, remains in the cells for a longer time in order to sustain the infection for a longer period. Such a condition can prolong the infection during pregnancy and consequently cross the placental barrier and infect the fetus.
