Microorganismos representam uma fonte promissora e relevante de compostos bioativos com aplicações farmacológicas e agrícolas. Dentre eles, os fungos endofíticos têm sido alvo de estudos que visam descobrir e explorar novos metabólitos e suas atividades, uma vez que exercem importantes papéis ecológicos e fisiológicos ao interagir com suas plantas hospedeiras durante, pelo menos, uma fase de seu ciclo de vida sem causar doenças. Dentre os critérios para a seleção da espécie hospedeira, destaca-se o uso medicinal. Fridericia platyphylla (Cham.) L.G. Lohman (Bignoniaceae) apresenta atividade antifúngica, tripanocida, leishmanicida e anti inflamatória. Tendo isso em vista, este trabalho tem como objetivo obter a diversidade de fungos endofíticos de Fridericia platyphylla, avaliar a citotoxicidade de seus extratos brutos frente a células tumorais de adenocarcinoma de cólon (HCT - 116) e identificar os componentes presentes em seus extratos. Para tal, os fungos endofíticos do indivíduo de F. platyphylla foram isolados e, após cultivo em BDA por 21, 28 e 60 dias, seus extratos brutos em acetato de etila foram triados por análise em CLAE, por potencial citotóxico (avaliado por meio do ensaio do MTT) e por rendimento. Diante desses resultados, cinco endófitos foram selecionados e identificados por sequenciamento da região ITS. Eles tiveram parte de seus metabólitos identificados por i) injeção de padrão e ii) por RMN após crescimento em larga escala. A injeção de padrão possibilitou identificar quatro endófitos produtores de alternariol, alternariol-9- metiléter, altenusina descarboxilada e altenusina, sendo dois deles identificados como Alternaria sp. A identificação por meio de RMN das substâncias isoladas resultantes do cultivo em larga escala do endófito Cercospora sp. relevou a presença de dois policetídeos: nova zelandina A e pirenocina A, sendo o primeiro relato da produção dos mesmos pelo gênero.

Microorganisms represent a promising and relevant source of bioactive compounds with pharmacological and agricultural applications. Among them, endophytic fungi have been the focus of studies aimed at discovering and exploring new metabolites and their activities, as they play important ecological and physiological roles by interacting with their host plants during at least one phase of their life cycle without causing diseases. Among the criteria for selecting the host species, medicinal use stands out. Fridericia platyphylla (Cham.) L.G. Lohman (Bignoniaceae) exhibits antifungal, trypanocidal, leishmanicidal, and anti-inflammatory activities. With this in mind, this work aims to obtain the diversity of endophytic fungi from Fridericia platyphylla, evaluate the cytotoxicity of their crude extracts against colon adenocarcinoma tumor cells (HCT-116), and identify the components present in their extracts. For this purpose, endophytic fungi from the individual F. platyphylla were isolated and, after cultivation on PDA for 21, 28, and 60 days, their crude extracts in ethyl acetate were screened through HPLC analysis, potential cytotoxicity (evaluated through the MTT assay), and yield. Based on these results, five endophytes were selected and identified by sequencing the ITS region. Some of their metabolites were identified through i) injection of standards and ii) NMR after large-scale growth. Injection of standardized metabolites allowed the identification of four endophytes producing alternariol, alternariol-9-methyl ether, decarboxylated altenusin, and altenusin, with two of them identified as Alternaria sp. Identification through NMR of isolated substances resulting from the large-scale cultivation of the endophyte Cercospora sp. revealed the presence of two polyketides: neozelandicin A and pyrenocin A, representing the first report of their production by the genus.
