Os tumores odontogênicos compreendem um grupo complexo de lesões de diversos tipos que podem criar dificuldades nos diagnósticos devido à sobreposição e diversidade de tipos histopatológicos e comportamentos clínicos. A compreensão do comportamento biológico dessas lesões é de fundamental importância para o diagnóstico final e planejamento do tratamento, pois influenciam o prognóstico. Dentre os tumores de origem epitelial odontogênica, o ameloblastoma é uma neoplasia benigna localmente invasiva, consistindo de epitélio odontogênico proliferativo, em meio a um estroma fibroso, não tendo potencial indutivo sobre o mesmo. É um tumor de crescimento lento, assimétrico, podendo provocar deformidade facial devido à distensão da cortical adjacente à lesão e nos casos mais avançados, romper o limite ósseo e aflorar na boca. Usualmente os ameloblastomas são diagnosticados na 4º e 5º décadas de vida, sendo que em 80 % dos casos, o tumor está na mandíbula e 20 % na maxila. O estudo da patogênese do ameloblastoma é dificultado devido as inúmeras vias de sinalização, diversidade de moléculas e as várias alterações genéticas que afetam o desenvolvimento e progressão do epitélio odontogênico presente no tumor. Este estudo tem como objetivo realização de analise imuno-histoquímica aos anticorpos das proteínas PTEN, mTOR, e NFkB a fim de identificar e correlacioná-las entre si nos casos de ameloblastomas. Foram selecionados 24 casos de ameloblastomas dos arquivos do Laboratório de Patologia Cirúrgica da Disciplina de Patologia Bucal da FOUSP para realização das analises imuno-histoquímicas. PTEN foi positivo em 29,2% das amostras, mTOR e NFkB foram positivos em 50% das amostras. Correlacionadas entre si, PTEN, mTOR e NFkB demonstraram positividade juntas em 8,3% das amostras e negativaram entre si juntas em 16,7% das amostras. Correlacionadas aos pares encontramos os seguintes valores de p no Teste do Qui Quadrado: PTEN x mTOR p=0.653; PTEN x NFkB p=0,653 e mTOR x NFkB p=0,414 concluindo que não existe associação significativa entre as proteínas.

Odontogenic tumors comprise a complex group of lesions of different types that can create diagnostic difficulties due to the overlap and diversity of histopathological types and clinical behaviors. Understanding the biological behavior of these lesions is of fundamental importance for the final diagnosis and treatment planning, as they influence the prognosis. Among the tumors of odontogenic epithelial origin, ameloblastoma is a benign neoplasm, but locally invasive, consisting of proliferative odontogenic epithelium, in the midst of a fibrous stroma, with no inductive potential over it. It is a slow-growing, asymmetrical tumor, which can cause facial deformity due to distention of the cortical bone adjacent to the lesion and, in more advanced cases, break the bone limit and emerge in the mouth. Ameloblastomas are usually diagnosed in the 4th and 5th decades of life, and in 80% of cases, the tumor is in the mandible and 20% in the maxilla. The study of the pathogenesis of ameloblastoma is difficult due to the numerous signaling pathways, diversity of molecules and the various genetic alterations that affect the development and progression of the odontogenic epithelium present in the tumor. The present study aims to perform immunohistochemical analysis of antibodies to PTEN, mTOR, and NFkB proteins in order to identify and correlate them with each other in cases of ameloblastomas. Twenty-four cases of ameloblastomas were selected from the files of the Laboratory of Surgical Pathology of the Discipline of Oral Pathology at FOUSP for immunohistochemical analyses. PTEN was positive in 29.2% of samples, mTOR and NFkB were positive in 50% of samples. Correlated with each other, PTEN, mTOR and NFkB showed positivity together in 8.3% of the samples and were negative together in 16.7% of the samples. In pairs, we found the following p-values in the Chi-Square Test: PTEN x mTOR p=0.653; PTEN x NFkB p=0.653 and mTOR x NFkB p=0.414, concluding that there is no significant association between the proteins.
