Recentemente foi evidenciado que o receptor nuclear rev-erb-α possui importante papel na adaptação da capacidade oxidativa do músculo esquelético, modulando a via de sinalização da AMPK. O exercício físico de endurance classicamente é capaz de ativar essa via de sinalização, porém os efeitos desse e de outros modelos de exercício físico (força e exaustivo) sobre a rev-erb-α permanecem desconhecidos. Além da regulação a nível transcricional dessa via de sinalização, já foi demonstrado que a rev-erb-α também é capaz de regular negativamente a autofagia no músculo esquelético e a biossíntese de dopamina no hipocampo. Sugere-se na literatura que a dopamina, juntamente com a serotonina, possui papel no desenvolvimento da fadiga cognitiva. Resumidamente, a rev-erb-α é candidata a modular as adaptações moleculares que medeiam o desempenho físico, fadiga do SNC e autofagia. Dessa maneira, a presente tese de doutorado tem como objetivo principal investigar o papel da rev-erb-α nas adaptações moleculares induzidas por diferentes modelos de exercício que regulam o desempenho físico, fadiga do SNC e autofagia. Os camundongos foram divididos em 4 grupos para o exercício agudo e 1 grupo controle (CT; sedentários): endurance (END; submetidos ao protocolo de exercício físico aeróbio contínuo), intervalado (INT; submetidos ao protocolo de exercício físico aeróbio intervalado), força (FOR; submetidos ao protocolo de exercício físico de força) e exaustivo (EX; submetidos ao protocolo de corrida até a exastão); 1 grupo para o exercício físico crônico - overtraining (OT; submetidos ao protocolo de overtraining) e 1 grupo controle - sedentários (SED). Os métodos utilizados foram: immunoblotting, RTqPCR e ELISA. Foi adotado o nível de significância de p<0,05 para os testes estatísticos utilizados. De maneira geral a Rev-erb-α foi extremamente sensível ao estímulo de exercício físico, inclusive a diferentes modelos, intensidades e volumes, tanto no músculo esquelético, quanto no hipocampo. Além disso, essa molécula apresentou adaptações em respostas ao overtraining, sugerindo que possa participar no desenvolvimento do estado de overreaching não-funcional (NFOR). A via da quinurenina (KYN) também foi sensível a sessão de exercício físico exaustiva, apresentando alterações para um perfil de produção de metabólitos neuro-tóxicos pelo gastrocnêmio. Interessantemente, foi evidenciada uma relação inédita entre a Rev-erb-α e a via da KYN. Portanto, a Rev-erb-α se apresenta como uma molécula importante e com várias atuações nas adaptações moleculares aos diferentes modelos de exercício físico.

Recently, the nuclear receptor rev-erb-α has been considered to play an important role in the skeletal muscle oxidative capacity adaptation, modulating the AMP-activated protein kinase (AMPK) signaling pathway. Classically, endurance physical exercise is able to activate this signaling pathway, but the effects of this and other physical exercise models (i.e., resistance and exhaustive) on rev-erb-α remain unknown. Besides the transcriptional regulation of this signaling pathway, rev-erb-α is also capable to regulate negatively the autophagy pathway in skeletal muscle and the dopamine biosynthesis in hippocampus. It is suggested in the literature that dopamine, together with serotonin, play a role in cognitive fatigue development. Briefly, rev-erb-α is candidate to modulate the molecular adaptations that regulate physical performance, CNS fatigue and autophagy. Thus, the main aim of the present thesis was to investigate the role of rev-erb-α in the molecular adaptations that regulate physical performance, CNS fatigue and autophagy after different exercise models. Mice were divided as follow: into 1 control group (CT, sedentary) and 4 groups to acute exercise endurance (END, submitted to continuous aerobic physical exercise protocol), interval (INT, submitted to interval aerobic exercise protocol), strength (FOR, submitted to strength physical exercise protocol) and exhaustive (EX; submitted to exhaustive running protocol); 1 group for chronic exercise training - overtraining (OT) and 1 sedentary group (SED). The following methodologies were used: immunoblotting, real-time polymerase chain reaction (qPCR) and ELISA. The level of significance of p < 0.05 was adopted. In general, Rev-erb-α was extremely sensitive to physical exercise stimuli, including different models, intensities and volumes, both in skeletal muscle and in the hippocampus. In addition, this molecule showed adaptations in responses to overtraining, suggesting that it may participate in the development of the NFOR state. The kynurenine (KYN) pathway was also sensitive to the exhaustive physical exercise session, showing a shift to a neurotoxic metabolite production profile in the gastrocnemius. Interestingly, an inedited relationship between Rev-erb-α and the KYN pathway was evidenced. Therefore, Rev-erb-α shows as an important molecule with several actions in molecular adaptations to different models of physical exercise.
