A resposta imune inata a bactérias do gênero Legionella vem sendo amplamente estudada nas últimas décadas, devido principalmente a grande quantidade de espécies do gênero capazes de provocar doença em humanos. Em países do sudeste asiático a Legionella longbeachae é extremamente relevante para a clínica por ser o agente etiológico de aproximadamente 50% dos casos de Legioneloses na região. Pouco se sabe sobre a resposta imunológica contra L. longbeachae, fato que se deve principalmente a recente descoberta da interação entre a bactéria e humanos, bem como a dificuldade da maioria dos países em detectar esta bactéria em pacientes, o que é crítico para a subnotificação de casos. Dentre os principais estudos realizados visando identificar componentes da imunidade inata relevantes para o controle da infecção, destacam-se dois trabalhos publicados na última década, um evidenciando o papel protetor do IFNγ e o outro, publicado por nosso grupo de pesquisa, apontando que as caspases inflamatórias 1 e 11 são dispensáveis para o controle da mortalidade causada por essa bactéria. No entanto, ainda não se sabe o que ocorre a jusante do engajamento do receptor de IFNγ, bem como outros componentes do sistema imune que operam na resposta contra a bactéria. Neste trabalho, nos demonstramos que a atividade do óxido nítrico está associada a uma redução da produção de IL-12 que leva a um agravamento da infecção por L. longbeachae. Adicionalmente, nossos estudos indicam que a expressão de NOS2 em neutrófilos de parece ser regulada pela morte celular piroptótica e que a piroptose, dependente de GSDMD é um acontecimento essencial para a imunidade à bactéria. Finalmente, mostramos que separadamente, os inflamassomas de AIM2 e NLRP3 são dispensáveis nesta resposta e que a clivagem de GSDMD em resposta a L. longbeachae ocorre mesmo na ausência de Caspase-1 e Caspase-11. Em conjunto, nossos dados sugerem que a morte celular dependente de GSDMD regula a produção de NO em neutrófilos para proteção do hospedeiro frente a infecção por L. longbeachae.

The innate immune response to bacteria of the genus Legionella has been widely studied in recent decades, mainly due to the large number of species of the genus capable of causing disease in humans. In Southeast Asian countries, Legionella longbeachae is extremely relevant to the clinic as it is the etiologic agent of approximately 50% of Legionellosis cases in the region. Little is known about the immune response against L. longbeachae, which is mainly due to the recent discovery of the interaction between the bacterium and humans, as well as the difficulty of most countries in detecting this bacterium in patients, which is critical for underreporting. of cases. Among the main studies carried out to identify components of innate immunity relevant to infection control, two studies published in the last decade stand out, one evidencing the protective role of IFNγ and the other, published by our research group, pointing out that inflammatory caspases-1 and 11are unnecessary to control the mortality caused by this bacterium. However, it is not yet known what happens downstream of the IFNγ receptor engagement, as well as other components of the immune system that operate in the response against the bacteria. In this work, we demonstrated that nitric oxide activity is associated with a reduction in IL-12 production that leads to an aggravation of L. longbeachae infection. Additionally, our studies indicate that NOS2 expression in neutrophils appears to be regulated by pyroptotic cell death and that GSDMD-dependent pyroptosis is an essential event for bacterial immunity. Finally, we show that separately, AIM2 and NLRP3 inflammasomes are dispensable in this response and that GSDMD cleavage in response to L. longbeachae occurs even in the absence of Caspase-1 and Caspase-11. Taken together, our data suggest that GSDMD-dependent cell death regulates NO production in neutrophils for host protection against L. longbeachae infection.
