O vírus Rocio, um Flavivirus brasileiro, produz epidemias com casos de infecção do sistema nervoso central humano que são muito graves, como foi evidenciado no Vale do Ribeira, SP, na década de 1970. Entretanto, achados soroepidemiológicos e encontros eventuais de indivíduos infectados com ROCV tem sido relatados, o que indica a circulação atual do vírus Rocio no Brasil, embora quase silenciosa. Há risco de re-emergência do vírus Rocio causando epidemias no Brasil. Esta Tese mostra como construímos um vírus Rocio repórter, com substância marcadora luminescente facilmente detectável. A construção dos vírus Rocio repórter foi feita expressando mCherry, mNeonGreen e HiBiT-tag. As construções foram feitas por Circular Polymerase Extension Reaction (CPER) onde fragmentos de DNA que perfazem todo o genoma viral foram submetidos a reação de circularização com fragmento Linker. Os genomas virais circularizados foram transfectados em cultura de células HEK 293T seguindo-se por infecção de células C6/36, que produziu sobrenadantes contendo vírus os quais foram coletados e armazenados. A expressão dos genes repórter para mCherry e mNeonGreen foram avaliadas por imunofluorescência, comprovando o resgate de vírus que expressaram estes genes repórter. Em suma, construímos com êxito vírus Rocio repórter contendo marcadores luminescentes e que deverão ter grande utilidade como marcadores da presença viral em muitos estudos futuros.

Rocio virus, a Brazilian Flavivirus, causes epidemics of severe human central nervous system infections, as demonstrated in the 1970s in the Ribeira Valley, SP. However, seroepidemiological findings and occasional encounters with individuals infected with ROCV have been reported, indicating that Rocio virus is currently circulating in Brazil, albeit almost silently. There is a risk that Rocio virus will reemerge and cause epidemics in Brazil. This work shows how we constructed a Rocio reporter virus with an easily detectable luminescent marker substance. The Rocio reporter viruses were constructed to express mCherry, mNeonGreen and HiBiT tag. The constructs were made using the circular polymerase extension reaction (CPER), in which DNA fragments forming the entire viral genome were subjected to a circularization reaction with a linker fragment. The circularized viral genomes were transfected into HEK 293T cell cultures, followed by infection of C6/36 cells, which produced virus-containing supernatants that were collected and stored. Expression of reporter genes for mCherry and mNeonGreen was assessed by immunofluorescence, demonstrating the rescue of viruses expressing these reporter genes. In conclusion, we have successfully constructed Rocio reporter viruses containing luminescent markers that should be very useful as markers of virus presence in many future studies.
