O inflamassoma é uma plataforma multiproteica formada a partir do reconhecimento de diferentes sinais, como o estresse ou dano celular e moléculas microbianas. Dentre os diferentes inflamassomas, NAIP/NLRC4 é um dos mais estudados e está envolvido no reconhecimento de moléculas microbianas no citosol, promovendo resistência do hospedeiro à infecção. Como objetivo principal do trabalho, propusemos identificar novas proteínas envolvidas na ativação do inflamassoma de NLRC4. Para isso, utilizamos a técnica de CRISPR-cas9 para deleção, em macrófagos, de genes previamente identificados em nosso laboratório, para avaliar seus possíveis papéis na ativação do inflamassoma. Dentre os genes, avaliados encontramos uma serina-treonina quinase que demonstrou ter um papel importante na via de ativação de NLRC4. Realizamos ensaios de replicação utilizando Legionella pneumophila como modelo, e descobrimos que a serina-treonina quinase tem papel importante no controle da infecção mediada por NAIP/NLRC4 tanto in vivo, quanto in vitro. Além disto, para investigar se este gene também estaria envolvido na ativação e morte celular por diferentes inflamassomas, macrófagos deficientes para a serinatreonina quinase foram gerados, e estimulados com nigericina, para avaliação do inflamassoma de NLRP3; poly (dA:dT) para avaliação de AIM2; Legionella pneumophila de tipo selvagem para avaliar NAIP/NLRC4; e L. pneumophila deficiente em flagelina para avaliar a ativação mediada via caspase-11. Nossos resultados demonstram que a serina-treonina quinase está envolvida na ativação de todos os inflamassomas, conforme observado pela redução da produção de IL-1β, caspase-1 e morte celular, além de uma redução na formação de punctas de ASC, indicando que essa proteína é importante para a oligomerização de ASC. Além disto, foi observado que a serina-treonina quinase colocaliza com os punctas de ASC, sugerindo uma interação física entre ela com o inflamassoma.

The inflammasome is a multiprotein platform formed from the recognition of different signals, such as stress or cellular damage and microbial molecules. Among the different inflammasomes, NAIP/NLRC4 is one of the most studied and is involved in the recognition of microbial molecules in the cytosol, promoting host resistance to infection. As the main objective of the work, we proposed to identify new proteins involved in the activation of the NLRC4 inflammasome. For this, we used the CRISPRcas9 technique for deletion, in macrophages, of genes previously identified in our laboratory, to evaluate their possible roles in inflammasome activation. Among the genes evaluated, we found a serine-threonine kinase that has been shown to play an important role in the NLRC4 activation pathway. We performed replication assays using Legionella pneumophila as a model, and found that serine-threonine kinase plays an important role in controlling NAIP/NLRC4-mediated infection both in vivo and in vitro. Furthermore, to investigate whether this gene is also involved in cell activation and death by different inflammasomes, serine-threonine kinase-deficient macrophages were generated, and stimulated with nigericin, to evaluate the NLRP3 inflammasome; poly (dA:dT) for AIM2 evaluation; wild-type L. pneumophila to assess NAIP/NLRC4; and flagellin-deficient L. pneumophila to assess caspase-11-mediated activation. Our results demonstrate that serine-threonine kinase is involved in the activation of all inflammasomes, as observed by reduced production of IL-1β, caspase-1 and cell death, in addition to a reduction in ASC puncta formation, indicating that this protein is important for ASC oligomerization. In addition, serinethreonine kinase was observed to colocalize with ASC punctas, suggesting a physical interaction between it and the inflammasome.
