Este estudo tem como objetivo avaliar o efeito de diferentes concentrações dos agentes antioxidantes - resveratrol e anetol - no metabolismo oxidativo de tecido ovariano bovino submetido ao cultivo in vitro. Os ovários bovinos foram fatiados e os fragmentos cultivados individualmente com 1000 µL de meio de cultura isolado ou suplementado. Vinte e quatro fragmentos foram removidos aleatoriamente e imediatamente fixados em paraformaldeído a 4% para o tratamento controle não cultivado (fresco). O meio básico (MB) para cultivo in vitro foi composto pelo meio essencial mínimo modificado (a-MEM) suplementado com BSA, ITS (insulina, transferrina, selênio), glutamina, hipoxantina, estreptomicina, penicilina e amicacina. Além disso, 336 fragmentos foram distribuídos aleatoriamente nos sete grupos de tratamento contendo MB ou MB suplementado com: resveratrol [1µM]; resveratrol [2µM]; resveratrol [5µM]; anetol [1000 µg/ml]; anetol [1500 µg/ml]; e anetol [2000µg/ml]. Todos os fragmentos foram avaliados através da histologia clássica (HE); microscopia confocal (MC) - sondas de iodeto de propídio (IP) (morte celular) e 2´7´-diclorodihidrofluoresceina (H2DCFDA) (estresse oxidativo); imuno-histoquímica (IH) - sondas FOXO3 (ativação folicular). Os dados são apresentados como média (±erro padrão da média) e percentagem. A probabilidade de p < 0,05 indica diferença significativa. O cultivo de tecido ovariano bovino leva à ativação massiva dos folículos no primeiro dia de cultivo, apesar da composição do meio. A suplementação com 2.000µg/mL de Anetol, embora boa para a ativação folicular, comprometeu a continuidade da viabilidade dos folículos. Por outro lado, o composto AN1500 não só preservou a viabilidade dos folículos, mas também promoveu a sua ativação.

This study aims to evaluate the effect of different concentrations of the antioxidant agents - resveratrol and anethole - on the oxidative metabolism of bovine ovarian tissue submitted to in vitro culture. Thus, bovine ovaries were sliced and the fragments were cultured individually with 1000 µL of culture medium alone or supplemented. Twenty-four fragments were removed randomly and immediately fixed in a 4% paraformaldehyde for the non-cultivated control treatment (fresh). The basic medium (BM) for in vitro culture was composed of the modified minimum essential medium (a-MEM) supplemented with BSA, ITS (insulin, transferrin, selenium), glutamine, hypoxanthine, streptomycin, penicillin, and amikacin. Furthermore, 336 fragments were randomly distributed in the seven treatment groups containing BM or BM supplemented with: resveratrol [1µM]; resveratrol [2µM]; resveratrol [5µM]; anethole [1000µg/ml]; anethole [1500µg/ml]; and anethole [2000µg/ml]. All fragments were evaluated through classical histology (HE); confocal microscopy (CM) - propidium iodide (IP) (apoptosis) and 2´7´-diclorodihidrofluoresceina (H2DCFDA) (oxidative stress) probes; immunohistochemistry (IH) - FOXO3 (follicular activation) probes. The data are presented as mean (± standard error of the mean) and percentage. The probability of p < 0.05 indicates a significant difference. The cultivation of bovine ovarian tissue leads to massive activation of follicles on the first day of culture, despite the medium composition. Supplementation with 2000µg/mL of Anethole, while being good for follicular activation, compromised the continued viability of the follicles. On the other hand, the AN1500 compound not only preserved the viability of the follicles, but also promoted their activation.
