O objetivo deste estudo foi explorar diferenças no perfil proteômico entre crianças e adolescentes divididos em grupos opostos de dano de DNA com diferentes nas concentrações sanguíneas de ácidos graxos poli-insaturados ômega-3 de cadeia longa. Metodologia: Meninos e meninas saudáveis (n = 140) de 9 a 13 anos de idade foram divididos com o teste k-means cluster em grupos opostos de dano de DNA, usando a variável tail intensity obtida por meio da técnica de eletroforese em gel de célula única (cluster 1 = 5,9% ± 1,2 e cluster 2 = 13,8% ± 3,1). Foram dosadas concentrações dos ácidos docosahexaenoico e eicosapentaenoico em eritrócitos e o perfil proteômico foi obtido pela SomaLogic Inc®. Resultados: O cluster de maior dano de DNA apresentou níveis mais baixos de DHA (5,4 ± 1,3, p = 0,003 / 6,2 ± 1,6 mg/dL) e EPA (0,5 ± 0,1 mg/dL, p <0,001 / 0,6 ± 0,2 mg/dL), e aumento da expressão das proteínas CDK8-CCNC, PIK3CA-PIK3R1, KYNU e PRKCB, que estão envolvidas em vias pró-inflamatórias. Conclusão: os resultados suportam a hipótese de que o papel protetor de ácidos graxos ômega-3 de cadeia longa contra o dano de DNA pode estar associado a expressão das proteínas CDK8-CCNC, PIK3CA-PIK3R1, KYNU e PRKCB.

The aim of this study was to explore differences in proteomic profiles and red blood cells levels of n-3 long chain PUFA that may be associated to DNA damage. Healthy boys and girls (n = 140) aged 9 to 13 years old in an urban area of Brazil were divided by k-means cluster test into two clusters of DNA damage (tail intensity) using the comet assay (cluster 1 = 5.9 % ± 1.2 and cluster 2 = 13.8 % ± 3.1). The cluster with higher DNA damage presented lower levels of DHA (6.2 ±1.6 mg/dL; 5.4 ±1.3, p = 0.003) and EPA (0.6 ±0.2 mg/dL; 0.5 ±0.1 mg/dL, p < 0.001), and increased expression of the proteins CDK8-CCNC, PIK3CA-PIK3R1, KYNU and PRKCB, which are involved in pro-inflammatory pathways. Our findings support the hypothesis that nutritional status regarding n-3 long chain PUFA have a protective role against DNA damage and that CDK8-CCNC, PIK3CA-PIK3R1, KYNU and PRKCB are differently expressed in clusters of DNA damage.
