Introdução: O processo de cicatrização cutânea consiste em um complexo evento que visa a restauração e homeostase teciduais. As úlceras crônicas constituem grande demanda no cotidiano clínico. A fototerapia por luzes de baixa potência (LASER) e não coerentes (LEDs) destacam-se como métodos bioestimuladores para o processo de cicatrização e reparo tecidual. Porém, encontramos poucas evidências cientificas sobre a associação da fototerapia por LASER e LED. Objetivo: avaliar a atuação da fototerapia LED Âmbar 590 nm e LASER Infravermelho 808 nm na cicatrização no modelo in vitro. Métodos: testes in vitro foram usados para avaliar a citotoxicidade da fototerapia isolada e associada em culturas de Fibroblastos NIH 3T3 e HaCaT (Queratinócitos humanos) pelo MTT(teste de citotoxicidade). Após o ensaio de scratch assay da fototerapia isolada e associada em fibroblastos e queratinócitos, dois grupos foram estabelecidos, sendo um submetido a estresse nutricional por baixa concentração de soro bovino fetal ( 2%) e outro sob condições normais (10%). Os resultados das culturas irradiadas com a fototerapia foram comparados aos das culturas não irradiadas. Foi padronizado um modelo de irradiação com fototerapia para os comprimentos de onda 590 nm e 808 nm de forma isolada e associada, seguindo todas as diretrizes atuais da WALT (Word association for photobiomodulation therapy). Resultados: O LED 590 nm isolado demonstrou melhor efeito na migração/proliferação no modelo do arranhão com queratinócitos HaCaT do que no modo associado. A fototerapia associada na cicatrização in vitro principalmente para célula HaCaT foi mais eficiente sobre as células em situação de estresse nutricional (soro bovino fetal a 2%). Conclusão: O comprimento de onda 590 nm (luz âmbar) estimulou migração/proliferação nas células irradiadas e demonstrou potencial de cicatrização principalmente nas culturas de queratinócitos tanto no modo isolado como associado.

Introduction: The cutaneous healing process is a complex event aimed at tissue restoration and homeostasis. Chronic ulcers are in great demand in daily clinical practice. Phototherapy by low power (LASER) and non-coherent (LEDs) lights stand out as biostimulating methods for the healing process and tissue repair. However, we found little scientific evidence on the association of LASER and LED phototherapy. Objective: to evaluate the performance of LED Amber 590 nm phototherapy and healing in the in vitro model. Methods: in vitro tests were used to evaluate the cytotoxicity of isolated and associated phototherapy in NIH 3T3 and HaCaT (Human Keratinocytes) Fibroblast cultures by MTT (cytotoxicity test). After the scratch assay of isolated and associated phototherapy in fibroblasts and keratinocytes, two groups were established, one submitted to nutritional stress due to a low concentration of fetal bovine serum (2%) and the other under normal conditions (10%). The results of cultures irradiated with phototherapy were compared to those of non-irradiated cultures. A model of irradiation with phototherapy was standardized for the wavelengths 590 nm and 808 nm in isolation and associated, following all current guidelines of the WALT (Word association for photobiomodulation therapy). Results: LED 590 nm alone showed better effect on migration/proliferation in the scratch model with HaCaT keratinocytes than in the associated mode. The associated phototherapy in in vitro healing mainly for HaCaT cells was more efficient on the cells under nutritional stress situation (fetal bovine serum at 2%) Conclusion: The wavelength 590 nm (amber light) stimulated migration/proliferation in irradiated cells and demonstrated healing potential mainly in keratinocyte cultures in both isolated and associated modes.
