A hidrocefalia é um desequilíbrio entre a produção, circulação e/ou absorção do líquido cefalorraquidiano, resultando em uma dilatação no sistema ventricular. O tratamento da hidrocefalia usualmente utilizado é cirúrgico (shunt), mas nem todos os pacientes podem ser submetidos ao tratamento imediatamente após o diagnóstico. Com isso, medidas neuroprotetoras vêm sendo testadas a fim de minimizar as lesões teciduais envolvidas. O canabidiol (CBD) se mostrou um composto com múltiplas ações neuroprotetoras pelo sistema nervoso central (SNC), como em: isquemias, doença de Alzheimer, efeito anti-inflamatório, entre outros. Dessa forma o objetivo deste trabalho foi avaliar o efeito do CBD na hidrocefalia experimental. Para estabelecer a dose utilizada, realizamos uma curva dose-resposta e a dosagem de 30 mg/kg/dia (intraperitoneal- ip.) foi escolhida e administrada em animais hidrocefálicos durante 21 dias. Foram utilizados ratos Wistar hannover submetidos à hidrocefalia através de injeção intracisternal de caulim, divididos em cinco grupos: controle intacto (n=10), hidrocefálicos sem tratamento (n=10), hidrocefálicos tratados com derivação (n=10), hidrocefálicos tratados com CBD ip. (30mg/kg/dia) (n=10) e hidrocefálicos tratados com derivação e CBD ip. (30mg/kg/dia) (n=10). Para avaliação do tratamento foram realizados testes de comportamento (open field, water maze e reconhecimento de objeto), estudos histológicos, imunoistoquímicos e bioquímicos. O tratamento do CBD reduziu a reação astrocitária (p<0,05) por imunomarcação do GFAP e pelo método de ELISA, redução da morte celular por apoptose no hipocampo (p<0,05) e melhorou a expressão de NeuN quando associados os dois tratamentos (p<0,05). Concluímos que o CBD apresenta efeitos benéficos quando associado ao tratamento cirúrgico da hidrocefalia experimental.

Hydrocephalus is an imbalance between the production, circulation and/or absorption of cerebrospinal fluid, resulting in dilation in the ventricular system. The treatment of hydrocephalus usually used is surgical (shunt), but not all patients can undergo treatment immediately after diagnosis. Thus, neuroprotective measures have been tested in order to minimize the tissue damage involved. Cannabidiol (CBD) proved to be a compound with multiple neuroprotective actions by the central nervous system, such as in: ischemia, Alzheimer's disease, anti-inflammatory effect, among others. Thus, the objective of this work was to evaluate the effect of cannabidiol on experimental hydrocephalus. To establish the dose used, we performed a dose-response curve and the dosage of 30 mg/kg/day (intraperitoneal-ip.) was chosen and administered to hydrocephalic animals for 21 days. Wistar Hannover rats submitted to hydrocephalus through intracisternal injection of kaolin were used, divided into five groups: intact control (n=10), untreated hydrocephalic (n=10), shunt-treated hydrocephalic (n=10), hydrocephalic treated with CBD ip. (30mg/kg/day) (n=10) and hydrocephalus treated with shunt and CBD ip. (30mg/kg/day) (n=10). To evaluate the treatment, behavior tests (open field, water maze and object recognition), histological, immunohistochemical and biochemical studies were performed. CBD treatment reduced the astrocytic reaction (p<0.05) by immunostaining of GFAP and by the ELISA method, reduced cell death by apoptosis in the hippocampus (p<0.05) and improved the expression of NeuN when associated with the two treatments (p<0.05) We concluded that CBD has beneficial effects when associated with the surgical treatment of experimental hydrocephalus.
