O vírus sincicial respiratório (RSV) é um dos patógenos de maior importância em saúde infantil, e até os 5 anos de idade, praticamente 100% das crianças já foram infectadas pelo menos uma vez pelo agente. Estudos do nosso grupo de pesquisa detectaram RSV em tonsilas de crianças assintomáticas e em linfonodos de pacientes adultos com hiperplasia linfoide benigna, sugerindo que RSV causa infecção prolongada e/ou persistente. É também conhecida a importância de IL-10 no estabelecimento de infecções virais persistentes. No presente estudo investigamos a persistência de RSV em modelo murino, comparando animais normais e knock out para IL-10. Camundongos C57BL6 wild type e knock out para o gene de IL-10 com 3 a 5 dias de vida foram infectados intranasalmente com 104 pfu de RSV num volume de 6,6 µL. Foram coletados pulmão, rim, cérebro, baço, timo e fígado nos tempos de 7, 15, 40, 80, 120 e 150 dias pós infecção. Encontramos genoma de RSV 150 dias após a infecção no fígado, timo, baço, rim e cérebro nos animais selvagens, mas não nos animais knock out para IL-10. Nestes últimos, aos 15 dias pós infecção não foi detectado vírus. A carga viral diminuiu ao longo do tempo, o que confirma o papel essencial de IL-10 para a persistência. Ambos os grupos, tanto o knock out para IL-10 quanto selvagem, os animais infectados apresentaram dano tecidual compatível com pneumonia e bronquiolite. Além disso, a expressão alterada de IFN γ na fase aguda nos animais sem IL-10 indicam o início da eliminação viral, compatível com a diminuição nas copias de genoma nesse grupo. Assim, o modelo murino de RSV torna-se útil para estudar mecanismos de persistência de RSV e o papel de IL-10 na infecção prolongada.

Respiratory syncytial virus (RSV) is one of the most important pathogens in child health, and by the age of 5, practically 100% of children have already been infected at least once by the agent. Studies from our research group detected RSV in asymptomatic children's tonsils and in lymph nodes from adult patients with benign lymphoid hyperplasia, suggesting that RSV causes prolonged and/or persistent infection. IL-10 is an important molecule in the establishment of persistent viral infections. In the present study we investigated the RSV persistence in a murine model, comparing wild type and knock out of IL-10 animals. C57BL6 wild type and knock out mice aged 3 to 5 days were infected intranasally with 104 pfu of RSV in a volume of 6.6 µL. The Lung, kidney, brain, spleen, thymus and liver were collected at 7, 15, 40, 80, 120 and 150 days post infection. We found RSV genome 150 days after infection in the liver, thymus, spleen, kidney, and brain in wild-type animals, but not in IL-10 knock out animals. In the latter, 15 days after infection no virus was detected. The viral load decreased over time, which confirms the essential role of IL-10 for persistence. In both groups, both IL-10 knock out and wild-type, infected animals showed tissue damage compatible with pneumonia and bronchiolitis. Furthermore, the altered expression of IFN γ in the acute phase in animals without IL-10 indicates the beginning of viral elimination, compatible with the decrease in genome copies found in this group. Thus, the RSV murine model becomes useful for studying mechanisms of RSV persistence and the role of IL-10 in prolonged infection.
